Embodiment

[0234] In this example, we increased endogenous PGE2 levels using a 15-PGDH inhibitor (SW033291), which inhibits the enzymatic function of 15-hydroxyprostaglandin dehydrogenase (15-PGDH), which Catalyzes the rate-limiting step in the catabolism of PGE2. Inhibition of 15-PGDH increases endogenous PGE2 and cAMP levels and enhances tissue repair in several mouse models of injury and disease. The present study demonstrates that elevating endogenous PGE2 levels through 15-PGDH inhibition results in renal vasodilation and confers renal protection against ischemic AKI.

Abstract

A method for preventing or treating renal ischemia reperfusion injury or acute kidney injury associated with renal ischemia reperfusion injury in a subject in need thereof includes administering to the subject a therapeutically effective amount of a 15-PGDH inhibitor.

Description

[0001] related application [0002] This application claims priority to U.S. Provisional Application No. 62 / 652,769, filed April 4, 2018, the subject matter of which is incorporated herein by reference in its entirety. Background technique [0003] Acute kidney injury (AKI) is an important clinical problem associated with high morbidity and mortality (1.7 million deaths per year). Considerable efforts have been made to develop preventive strategies for AKI using a variety of agents and animal models. Despite advances in prevention strategies, no specific treatment for AKI has been developed. [0004] The main etiology of AKI is hypoxia and oxidative stress caused by renal ischemia-reperfusion injury (IRI). Inadequate oxygen supply during transient decreases in renal blood flow (RBF) can lead to impaired extrarenal medulla energy (ATP depletion), which in turn leads to renal tubular epithelial cell damage and death due to acute tubular necrosis (ATN) and apoptosis. Inflamma...
