Problems solved by technology

[0006] Therefore, it is necessary to provide a method that can quickly and accurately distinguish between viable and non-viable SARS-CoV-2, which will be very important supporting information for doctors managing Covid-19 patients, because cell culture systems and animal models have not been shown to be reliable or practical

Embodiment approach

[0017] Some typical embodiments embodying the features and advantages of the present application will be described in detail in the description in the following paragraphs. It should be understood that this case can have various changes in different aspects without departing from the scope of this case, and the descriptions and drawings therein are used for illustration in nature rather than to limit this case.

[0018] The present invention develops a method for the identification of infectious and inactivated / killed viruses in clinical samples, thereby providing clinical diagnostic information for the management and treatment of Covid-19 patients. The method uses a nucleic acid intercalating dye or chemical that selectively enters cells with damaged membranes, while intact membranes provide a barrier to the molecule. Once inside a (dead) cell, the dye or chemical is embedded in the nucleic acid of the cell and covalently cross-links with the nucleic acid after exposure to st...

Abstract

The invention provides a method for detecting human coronavirus infectivity. The method comprises the following steps: (a) dividing a sample to be detected into a first sample and a second sample; (b) treating the first sample with an intercalation dye or chemical reagent; (c) exposing the first sample to light for photoactivation; (d) amplifying the target nucleic acid in the first sample and the second sample; and (e) determining the infectivity of the human coronavirus according to the amplification results of the first sample and the second sample.

Description

【Technical field】 [0001] This case concerns the diagnosis of a human coronavirus, especially a method for detecting the infectivity of a human coronavirus. 【Background technique】 [0002] Traditional culture and microscopy methods for detecting living cells are tedious, laborious and time consuming. There are also methods that can assess viability through staining techniques, such as BacLight fluorescence microscopy or acridine orange, or flow cytometry with conjugated dyes, and cellular respiration physiological tests that cannot detect specific pathogen species. These culture-based methods present some challenges, such as isolation and identification of specific pathogens in a large background microbiota, detection of low-level pathogens, and potential biosafety for operators and the environment during tedious handling question. In addition, these culture-based methods suffer from another problem, that is, some human pathogens may enter a "viable but non-culturable (VBNC...
