Embodiment 1

[0042] Embodiment 1: Production protease HY-3

[0043] For the production of protease HY-3, the standard medium used for culturing microorganisms was sterilized at 121° C. for 20 minutes under high pressure, and then Aranicola proteolyticus HY, which produces protease HY-3, was added in an amount of 0.1-5% by volume based on the total volume of the medium -3 strain (KCTC 0268BP) and cultivated at 25-30°C for 25-30 hours. The medium is subjected to membrane filtration, which separates the supernatant from the biomass. When necessary, the supernatant was concentrated 3-10 fold by filtration through a 10 kDa membrane.

Embodiment 2

[0044] Example 2: Analysis of proteins eluted from liming and deliming steps using protease HY-3

[0045] In order to study the role of protease HY-3 in the liming and deliming steps which greatly affect COD, the following procedure was carried out.

[0046] Specifically, 20 g of raw hide was cut into an appropriate size, put into a 50 ml test tube, and then 10 ml of water was added thereto. Thereafter, 0.5% sodium bisulfite and 0.5% ammonium sulfate were added and reacted at room temperature for 15 minutes. After the reaction, 0.2% detergent and 0.5% degreaser were added to the resulting solution and reacted at room temperature for 30 minutes. Three tubes were divided into a control containing no protease HY-3, and containing the supernatant prepared as in Example 1 above and the concentrated supernatant. Allow each tube to stand at room temperature and obtain a sample. The protein analysis kit (Bio-rad, USA) modified from the Bradford method (Bradford, M., Anal. Biochem.,...

Embodiment 3

[0048] Example 3: Use of protease HY-3 in the softening step

[0049] In order to improve the stability and performance of the protease HY-3 in leather processing, the protease was formulated and thus used in the softening step.

[0050] The lyophilized protease HY-3 of the present invention is added to 40-50% ammonium chloride (NH 4 Cl), 40-50% ammonium sulfate ((NH 4 ) 2 SO 4 ), 0.005-0.01% calcium chloride (CaCl 3 ) and 0.025-0.1% lactose. In order to examine the softening effect of the formulated protease HY-3, hides were subjected to a deliming step followed by a softening step using different enzyme preparations, after which the surface state of the hides was observed. In addition, hides and skins are subjected to tanning and finishing steps, and then the surface state is observed. As protease preparations used in the softening step for comparison, Amoron (Chungmu Fermentation, Korea) and Oropon K (TFL, Germany) were used. The experimental procedure is summarized ...

Abstract

The present invention relates to a method for preparing leather using a protease and a method for treating wastes derived from leather processing using the same, which are advantageous in terms of preparation of leather of excellent quality, decreased waste production by reducing the amount of chemicals, and treatment or recycling of wastes in an environmentally friendly manner. The protease HY-3 produced from Aranicola proteolyticus HY-3 strain is added to the steps of soaking, liming, deliming and bating, among leather processing, and thus the leather is prepared. Additionally, the protease HY-3 is added to wastewater and solid wastes generated during the soaking and liming steps, and solid wastes generated during the liming, deliming, bating and finishing steps, so that the wastes are treated.

Description

technical field [0001] The present invention relates to a process for the preparation of leather using protease HY-3 and a process for using it to treat waste from leather processing. More specifically, the present invention relates to a method for preparing leather, wherein protease HY-3 produced from Aranicola proteolyticus HY-3 strain is added to the soaking, liming, deliming and softening steps; Process for waste water and solid waste from soaking and liming steps, and solid waste from liming, deliming, softening and finishing steps, which has the advantage of producing leather of superior quality and environmentally friendly treatment or recycling of waste . Background technique [0002] In general, the leather industry is a chief industry that determines the quality of leather products such as shoes, bags, garments, and belts as a rough leather processing industry requiring expensive equipment and technology. In the leather industry, when hides or hides of various an...
