Embodiment 1

[0082] Example 1: Ability of Valerian Extract to Bind Strongly to GABA Receptors

[0083] Valerian, a medicinal herb that produces anxiolytic and sedative effects, is known to bind to GABA receptors and thus potentiate the sedative effects of anesthetics and other drugs that act on GABA receptors. See Yuan, Chun-Su et al., 2004. "The Gamma-Aminobutryic Acidergic Effects of Valerian and Valerenic Acid on Rat Brainstem Neuronal Activity." Anesth. Analg. 98:353-8.

[0084] Those of ordinary skill in the art know that there are many methods for determining the ability of a substance to bind to GABA receptors. An example of this approach is set forth below and in Yuan, Chun-Su et al., 2004. "The Gamma-Aminobutryic Acidergic Effects of Valerian and Valerenic Acid on Rat Brainstem Neuronal Activity." Anesth. AnaIg. 98: 353-8 description, the entire contents of which are incorporated by reference herein. Yuan et al. identified that the anxiolytic and sedative effects of valerian are...

Embodiment 2

[0093] Embodiment 2: the extraction of Radix Paeoniae Alba

[0094] Native Radix Paeoniae Alba root, without pretreatment or excipients, was ground into powder and the Radix Paeoniae Alba powder was refluxed with aqueous 5% methanol at about 80-84°C for 3 hours.

Embodiment 3

[0096] Embodiment 3: TLC analysis of Radix Paeoniae Alba root extract

[0097] Thin layer chromatography (TLC) plates were available in 20 x 20 cm and 1000 micron format, coated with a prefabricated layer of UV grade 254 silica gel (catalogue #08002) from Analtech, Inc. (Newark, DE). TLC plates were coated with a mobile phase containing methanol:chloroform:ethyl acetate (v:v:v) in a ratio of 1:2:1.

[0098] TLC plates were coated with the solution in Example 2 described above and dried. Different rings on the TLC plate can be detected visually at UV 254 nm and the scraped TLC plate collected. Separation was repeated by TLC until pure fractions (>92% paeoniflorin) were obtained. Individual fractions were then analyzed by HPLC.

Abstract

The present invention relates to medicine composition for treating insomnia. The medicine composition includes white peony root extract, especially paeoniflorin, and at least one of date extract, polygala root extract and passion flower extract. The medicine composition is combined with GABA receptor to reach the synergistic effect of improving sleep quality.

Description

[0001] This application claims priority and is a continuation-in-part of US Patent Application 11 / 075,010 filed March 7,2005. Background technique [0002] Insomnia or insomnia is a very common condition, usually caused by excessive mental and / or physical stimulation. For example, stress, poor eating habits, lack of physical activity, and psychological effects are the main factors that cause insomnia. Insomnia also often reflects a lack of central inhibitory processes. For example, gamma-aminobutyric acid (GABA) is the major inhibitory neurotransmitter of the central nervous system (CNS). Several studies have shown that activation of GABA receptors, by binding to GABA, contributes to sleep. Additionally, during times of stress and anxiety, the body produces less GABA, making it more difficult for the body to calm down due to the reduced inhibition of GABA by binding to GABA receptors. The result is often insomnia. Inability to sleep is an important problem because sleep is...
