Embodiment Construction

[0025] Below in conjunction with accompanying drawing, further describe the specific implementation of the present invention in detail with enrichment and separation of microorganisms in the deep sea sediment sample:

[0026] (1) Extraction of the total genome in the sample

[0027] (1) Under sterile conditions, weigh 3g of the deep-sea sediment sample and transfer it into a sterile centrifuge tube with a volume of 50ml, add 30ml of sterile artificial seawater basic salt, shake fully and place it on a shaker at a constant speed of 180rpm Shake for 10-20 minutes, then centrifuge at 3000rpm for 3 minutes, transfer the supernatant to a new centrifuge tube; wash the precipitate twice more, collect the supernatant, and save the precipitate for later use.

[0028] (2) The supernatant was centrifuged at 12000rpm for 15min to collect the precipitate.

[0029] (3) Add 20ml of sterilized phosphate buffer solution to the precipitate, shake thoroughly, place on a shaker at a constant spe...

Abstract

The invention discloses a process for cultivating and screening microbe strain, which comprises firstly extracting general genome of each sample, the detecting the kind, abundance, biodiversity and distribution rule of microbe in samples through utilizing the PCR technology and the RFLP molecular biology technology, analyzing the components of mineral elements in sample through using an energy spectrometer, building an individual enrichment culture system for various microbe in sample through combining physicochemical information of sampling points which is obtained in sampling, and screening various microbe in separating sample, especially target microbe using the individual enrichment culture system. Compared with the traditional selective microbe screening culture process, the process changes some microbe which can not be cultivated into microbe which can be cultivated, overcomes the problems of larger new species loss probability, large sample consumption loss, high randomness and blindness degree and low efficiency, and increases the yield and kind of microbe which is cultivated and bred from sample.

Description

technical field [0001] The invention relates to a screening method for microbial strains, in particular to a method for screening, separating and cultivating microbial strains in extreme environment samples. Background technique [0002] Microbes, especially those in extreme environments (such as acid / alkali / temperature / cold / salt samples, deep-sea samples, crater samples, high-altitude samples, space samples, etc.), play an extremely important role in natural biology, geochemical cycles, etc. character of. The study of microbes in extreme environments not only helps us to understand and understand the origin and evolution of life, the tolerance mechanism of extreme environments, and the interaction between them and the environment; but also their gene resources, enzyme resources and their metabolite resource industries are known as A sunrise industry with sustainable development in the 21st century. The competition for microbial strain resources, especially those with spec...
