Embodiment 1

[0027] a) 97 F. 1 group.

[0028] b) Analyze the two parents and their F 1 Population, statistical analysis of genetic types of polymorphic sites between parents in offspring populations. A total of 732 polymorphic marker loci, including 23 SSRs, S gene loci, 226 SRAPs, and 482 AFLPs, were finally obtained, and then linkage analysis was performed on them using Jionmap3.0 mapping software to construct a table containing Genetic linkage map of 240 polymorphic markers in 'August Red' pear.

[0029] Using SSR, SRAP, and AFLP molecular marker methods, the 'August Red' and 'Dangshan Suli' and their offspring were analyzed, and the genetic types of the polymorphic sites between the parents were statistically analyzed in the offspring population.

[0030] For the experimental procedure of SSR labeling, refer to the method of Yomamoto T. et al. (Euphytica, 2002, 124:129-137); for the experimental procedure of SRAP labeling, refer to the method of Li et al. (Theor Appl Genet, 2001, 1...

Embodiment 2

[0042] Using the screened molecular markers linked to the main QTL for fruit soluble sugar content of 'Augusthong', the 97 F strains of 'Augusthong' and 'Dangshansuli' were compared 1 Preliminary testing was carried out on the offspring of the hybrid to test the practical value of this method in molecular marker-assisted selection of soluble sugar content in pear fruit. Use the genomic DNA of the 97 offspring and SRAP primers me6 and pm19 to carry out PCR reactions. After electrophoresis, determine whether there is a corresponding marker according to the presence or absence of the me6pm19-1300 band. If there is a marker, it means that the fruit of the plant has a high content of soluble sugar. , if it does not exist, it means low. At the same time, it is verified by the actual measured fruit soluble sugar content and molecular marker detection results.

[0043] The results showed that among the 97 F1 hybrid progenies of 'August Red' and 'Dangshansuli', a total of 51 strains sh...

Abstract

The invention relates to a molecular marker for the content of soluble sugar in an 'August red' pear fruit, which belongs to the field of inheritance breeding. A genetic linkage map of the 'August red' pear fruit is constructed by using SRAP (Sequence-Related Amplified Polymorphism), SSR (Simple Sequence Repeats) and AFLP (Amplified Fragment Length Polymorphism), a colony is analyzed by using an interval mapping method in combination with phenotypic identification of the content of the soluble sugar in the fruit, and the existence of major QTL (Quantitative Trait Loci) on a seventh linkage group of the 'August red' pear fruit is detected, so that the characteristic of the soluble sugar content of 12.4 percent can be explained. A marker which is closest to the locus Psg-1 of the major QTL is me6pm19-1300, wherein the distance is 0 centimeter. The obtained major QTL molecular marker of the soluble sugar plays an important theoretical and practical guide role in accelerating the genetic improvement process of a pear variety and improving the breeding selection efficiency.

Description

1. Technical field [0001] The invention provides a molecular marker for the soluble sugar content of August red pear fruit, belongs to the field of molecular genetic breeding, and can be used for early molecular assisted selection of the soluble sugar content traits of pear fruit to improve breeding efficiency. 2. Background technology [0002] pear( Pyrus L.) is native to my country and is the third largest fruit tree species in my country. Pears are deeply loved by consumers because of their high nutritional value, sweetness and juiciness. Soluble sugar content is one of the important factors affecting the quality and flavor of pear fruit, and is the primary factor determining the sweetness and taste. Consumers in my country generally like pears with high sweetness, so cultivating varieties with high soluble sugar content has become a pear breeding strategy. one of the important goals. [0003] Since pear is a perennial woody fruit tree crop, most of the traits relate...
