Abstract

Provided are a method for constructing an MHC region nucleic acid library, an MHC region nucleic acid library, a method for sequencing the MHC region, a method for determining a type of gene mutation in the MHC region, and a use of the method for sequencing the MHC region in MHC genotyping. The method for constructing an MHC region nucleic acid library comprises the following steps: preparing a genomic library, wherein the genomic library comprises genomic DNA segments; and screening the genomic library by using a probe to obtain a target library, wherein the probe is specific for the MHC region, and the target library forms the MHC region nucleic acid library.

Description

technical field [0001] The present invention relates to a method for constructing a nucleic acid library, in particular to a method for constructing a target library, especially a nucleic acid library for the MHC region, and its application. Background technique [0002] Major Histocompatibility Complex (MHC) refers to a group of tightly linked gene groups on a chromosome of vertebrates that encode major histocompatibility antigens, control mutual recognition between cells, and regulate immune responses. The discovery became one of the most studied regions of the vertebrate genome after it was discovered in mice in 1936. [0003] Human major histocompatibility antigen was first discovered on the surface of leukocytes, so it is called human leukocyte antigen (human leucocyte antigen, HLA) [1] . The HLA complex is located at 6P21.31, the short arm of human chromosome 6, with a total length of about 4MB. From the centromere side, it is located in the regions of class II genes...
