Embodiment Construction

[0017] The present invention will be further described in detail through the following examples in conjunction with the accompanying drawings, but the scope of the present invention will not be limited in any way.

[0018] In this example, the specific high expression of the ZmPEP1 gene in young maize heart leaves was verified by real-time fluorescent quantitative PCR (FQ-PCR), thereby cloning its promoter, which is 1976bp in length, and its sequence is shown in SEQ ID No in the sequence listing. :1.

Abstract

The invention discloses a promoter of the specificity of heart leaves of Zea mays ssp. Mays L. and an application thereof. The promoter which is a promoter of a gene ZmPEP1 of Zea mays ssp. Mays L. has a nucleotide sequence represented by SEQ ID No:1 or a nucleotide sequence which can be hybridized with the nucleotide sequence represented by SEQ ID NO:1 under strict conditions and has same functions with the nucleotide sequence represented by SEQ ID NO:1. The promoter of the present invention, which has the specificity of heart tissues, can be applied to the field of transgenic plant breeding. By constructing a plant expression vector containing the promoter, the promoter is converted to plants (such as Zea mays ssp. Mays L.) to drive downstream genes to specifically express in the heart leaves of the plants, so the breeding purpose is reached.

Description

technical field [0001] The invention belongs to the technical field of plant genetic engineering and relates to the cloning of a plant promoter and its application, in particular to the cloning of a corn heart-leaf specific promoter and its function analysis. Background technique [0002] Corn is not only the main crop for food and feed, but also an important industrial raw material. With the popularization and acceleration of industrialization, the contradictions between the decrease of arable land, the deterioration of the environment and the shortage of food have become increasingly acute, while conventional breeding techniques cannot meet human needs for high-yield, high-quality, disease-resistant and stress-resistant new corn varieties. For more than ten years, a batch of new varieties of corn that are insect-resistant, disease-resistant, herbicide-resistant, salt-resistant, drought-resistant, and high-quality have been bred using transgenic technology. [0003] Corn b...
