Problems solved by technology

However, due to the presence of various mycotoxins in the Trichoderma fermentation product, and the low activity of cellulase, especially the low activity of β-glucosidase, cellobiose accumulates in the reaction system and affects the enzymatic hydrolysis efficiency. Its scope of application is limited

Abstract

The present invention discloses a gene encoding alkaline beta-glucosidase and application thereof. A gene Pbgl of the alkaline beta-glucosidase contains a nucleotide sequence of SEQ ID NO. 1; and an amino acid sequence of the said gene encoding the alkaline beta-glucosidase is provided, and shown as a SEQ ID NO. 2. Activity of the alkaline beta-glucosidase adapts a pH range of 5-10, and an optimum pH is 8.0. When pH is 10, enzymatic activity still retains at 58% of the highest activity, and is superior to the activity of an existing alkaline beta-glucosidase. Therefore, the alkaline beta-glucosidase has improved application value, and is suitable for application to the decomposition of cellooligosaccharide.

Description

technical field [0001] The invention belongs to the technical field of genetic engineering and enzyme engineering, and relates to a gene encoding alkaline β-glucosidase and application thereof. Background technique [0002] β-glucosidase (beta-glucosidase, EC3.2.1.21), also known as β-D-glucoside glucohydrolase, widely exists in various organisms from microorganisms to vertebrates. It can hydrolyze terminal non-reducing β-D-glucosidic bonds such as salicin, cellobiose, cellotriose, cellotetrose and p-nitrobenzene -β-D-glucoside (pNPG), etc., play an important role in the release of food flavor substances and cellulose degradation (SesteloA.B.F, Poza M., Villa T.C.β-Glucosidase activity in a Lactobacillusplantarum wine strain.World Journal of Microbiology & Biotechnology, 2004, 20: 633-637). [0003] Energy shortage is one of the major problems facing this century. As the main polysaccharide product of plant photosynthesis, cellulose is the most abundant carbohydrate on ea...
