The method includes: measuring total nitrogen TN0, total phosphorus TP0 and BOD5 in wastewater to be treated; determining effluent total nitrogen TNS and total phosphorus TPS required in emission standard to calculate theoretical carbon source demand CS according to the emission standard executed by sewage treatment plants. The ratio of the theoretical carbon source demand CS to wastewater BOD5 concentration is DNR (difficulty of nitrogen removal) in biological nitrogen and phosphorus removal in wastewater treatment. The method can be used to quickly estimate carbon source supply for wastewater treatment so as to determine the difficulty of nitrogen and phosphorus removal for wastewater according to quality parameters of conventional inflow. In practical operations by sewage treatment plants, corresponding treatment schemes are determined according to the estimation results.
