[0020] (1) Amplify the nucleotide fragment containing the SNP site

[0021] Primers were designed according to the IFITM3-rs12252 gene sequence (SEQ ID NO.1), including the forward primer F5'-GAAAAGGAAACTGTTGAGAACCGAA-3' and the reverse primer R5'-GAGCCTCCTCCTAAACCTGCAC-3' to amplify the nucleotide of the SNP to be tested fragment. The SNP site is located at 142bp of the IFITM3-rs12252 gene sequence, and the base at this position can be C or T. When the base at this position is T, it can be recognized by MscI endonuclease (the recognition sequence is TGG^CCA).

Abstract

The invention provides an SNP (Single Nucleotide Polymorphism) marker related to human severe influenza and an application thereof. The SNP marker related to human severe influenza is located at 142bp of the sequence of the IFITM3-rs12252 gene shown in SEQ ID NO. 1, and a basic group at 142bp is a C individual, which is at higher risk of suffering from severe influenza than the base group at 142bp, which is a T individual. Polymerase chain reaction (PCR) and sequencing technique are used for screening the SNP marker related to human severe influenza, a restricted fragment length polymorphism (RFLP) method is used for detecting the genotypes of a crowd to be tested, and the early prediction of severe influenza and the control on the generation of severe influenza are possible according to genotype judgment results. In addition, a genotype detecting method provided by the invention avoids the investment of expensive instrument, has the advantages of simpleness, rapidness, low cost and the like and is convenient for popularization and application at a basic level.

Description

technical field [0001] The invention relates to the fields of genetic engineering and molecular biology, in particular to a SNP marker in the IFITM3-rs12252 gene related to human severe influenza and its application. Background technique [0002] Influenza (Influenza) is highly contagious and has a high incidence rate, which can easily cause outbreaks and seriously threaten human health. Although influenza vaccine can effectively prevent the occurrence of influenza, the effect of influenza vaccine can be weakened after the mutation of influenza virus. For example, the H1N1 influenza outbreak in 2009, according to statistics, among the 127,000 patients diagnosed in my country, 8,320 were severe and critical cases, and 196 died. It can be seen that influenza, especially severe influenza, has become a major public health problem that threatens human health. [0003] Interferon-induced transmembrane protein 3 (IFITM3), located on human chromosome 11, is a recently discovered p...
