[0047] The aim of this study was to identify the extent of ERG rearrangements in PIN lesions of the prostate. The inventors developed an automated staining protocol to detect ERG overexpression by immunohistochemistry (IHC) in combination with detection of specific gene rearrangements by FISH assay. To this end, probes specific for ERG gene rearrangements comprising 3' and 5' ERGs were detected with quantum dot (Life Technologies, OR) bioconjugates. Fixatives commonly used for needle biopsy preservation and the duration of fixation were also assessed. Specific FISH signals were deconvoluted using an interferometer and spectral imaging software.
