Abstract

The invention discloses a new application of a transcription factor DksA gene. The dksA gene in Photorhabdus luminescens LN2 plays an important role in regulating the pigment absorption of P.luminescens LN2 colony, the production of fluorescent protein of P.luminescens LN2, the stickiness of P.luminescens LN2 colony, the toxicity of P.luminescens LN2 to the wax moth, P. The virulence of luminescens LN2 to the nematode Heterorhabditis bacteriophora H06, and the application of P.luminescens LN2 to the recovery of H.indica LN2 infected nematodes. The dksA mutation had obvious effects on the physiological and biochemical characteristics of the symbiotic bacterium LN2. The mutant strain LN2△dksA significantly delayed the recovery and oviposition of nematodes during the LN2 infection stage; lost the toxicity to the non-specific symbiotic nematode H.bacteriophora H06, and could support the growth and reproduction of the H06 nematode; the injection toxicity to the wax moth was significantly reduced ; but did not significantly affect the growth of symbiotic bacteria and the in vitro culture yield of LN2 nematodes.
