Problems solved by technology

[0006] At present, there is no research on the genetic variation of the Notch1 gene in cattle at home and abroad, because the function of the Notch1 gene involves important growth traits such as muscle differentiation and development.

Embodiment Construction

[0029] The present invention will be further described below in conjunction with the accompanying drawings and embodiments, so that the public can have an overall and sufficient understanding of the content of the invention, rather than limiting the protection scope of the present invention.

[0030] The present invention designs primers according to the sequence of the NICD region of the Notch1 gene, uses the DNA pool containing 50 Qinchuan cattle genome samples as a template, and performs PCR amplification. After sequencing, it is found that there is a single nucleotide polymorphism at position 48250 of the Notch1 gene, see figure 1 . A missense codon mutation was formed when the A at position 48250 of the Notch1 gene was mutated to G, that is, the amino acid at position 13780 was mutated from His to Arg. For the above SNP site, the present invention can detect its single nucleotide polymorphism simply, quickly, at low cost and accurately by designing primers for PCR amplifi...

Abstract

The invention discloses a method and a kit for detecting RFLP of an SNP site of a cattle Notch1 gene. The method comprises the following steps: with a cattle blood whole genome DNA as a template, carrying out PCR amplification on a segment of sequence in an NICD region of the cattle Notch1 gene; digesting a PCR amplification product with a restriction enzyme HhaI; carrying out agarose gel electrophoresis on the segment after enzyme digestion; and identifying single nucleotide polymorphism of the NICD region of the Notch1 gene according to the agarose gel electrophoresis result. The SNP is arranged in the NICD region with an important function, of the Notch1 gene, and is related to a plurality of important economic traits of the cattle. A foundation is laid for building the relationship between the SNP of the Notch1 gene and growth traits; and the detection method disclosed by the invention can be applied to marker assistant selection of the growth traits of the cattle, thus a cattle population with excellent genetic resources is rapidly built.

Description

technical field [0001] The invention belongs to the field of molecular genetics, in particular to rapid detection of the single nucleotide polymorphism (SNP) of Notch transmembrane receptor family 1 gene based on RFLP (restriction fragment length polymorphism). Background technique [0002] Single nucleotide polymorphism (SNP) refers to the polymorphism caused by the substitution of a single nucleotide (A / T / C / G) in the genomic DNA sequence. SNP belongs to the third-generation molecular marker, which has the advantages of high density, biallelic genes, and easy automatic detection. Because SNP has advantages that other markers cannot match, it has been widely used as a research tool in various fields of life sciences. application. [0003] In recent years, people have developed many methods for searching molecular genetic markers, the most common ones are single-strand conformation polymorphism (SSCP), direct sequencing technology and PCR-RFLP, etc. Subjective factors have ...
