Embodiment 1

[0029] 1. Test object (test material)

[0030] The bream used in the experiment was first-year-old bream, with a total of 1500. The samples were collected from Hubei Bairong Aquatic Fine Breed Co., Ltd. After being salvaged from the pond, it was temporarily raised in a bucket until it was stable, and then injected with Aeromonas hydrophila to infect it. The concentration of injected Aeromonas hydrophila was 1×10 7 cfu / mL, the water temperature was kept at 27-29°C. Fin ray samples from fish that died within 12 hours (susceptible group) and survived after 5 days (resistant group) were collected, stored in absolute ethanol, and then brought back to the laboratory for storage at -20°C.

Abstract

The invention belongs to the technical field of fish molecular mark screening and application and particularly relates to a megalobrama amblycephala transferrin receptor gene SNP molecule mark and applications thereof. The molecule mark can be used for assisting megalobrama amblycephala disease-resistant variety selection. The molecule mark is prepared by cloning from a transferrin receptor gene that is GenBnk accession No.KM225267. The nucleotide sequence of the molecular mark is shown as SEQ ID NO:1. The R at the number 267 base site of the sequence is A or G (with the mutant being in a gene intron portion). The mutant leads to NocI enzyme digestion polymorphism. Application of utilizing the screened molecular mark to perform correlation analysis on megalobrama amblycephala disease-resistant characters proves that the molecular mark can be used for assisting megalobrama amblycephala disease-resistant character selection.

Description

technical field [0001] The invention belongs to the field of preparation of fish molecular markers, and in particular relates to a SNP molecular marker of transferrin receptor gene of bream and its application. The molecular marker can be used to assist the breeding of disease-resistant varieties of bream. Background technique [0002] Megalobrama amblycephala (Megalobrama amblycephala), also known as Wuchang fish, belongs to Cyprinidae, Cyprinidae, Cyprinidae, and Bramy genus. It is an important economically farmed fish in my country. With the promotion of pomace bream farming across the country, pomace pomace gradually occupies a certain proportion in the production of freshwater aquaculture fish. In recent years, the increase in people's demand for bream has led to further expansion of the breeding scale, and bream has become one of the main economically farmed fish in my country. However, in the process of large-scale farming, due to inbreeding, high breeding density, a...
