Abstract

The invention relates to a dCAPS (Derived Cleaved Amplified Polymorphic Sequence) marker development method for identifying flooding tolerance of chrysanthemum and application, which belong to the technical field of biology. The method comprises the following steps of a, digging SNP (Single Nucleotide Polymorphism) loci remarkably related to the flooding tolerance of the chrysanthemum through a GWAS (Genome Wide Association Study) method; b, analyzing SNP mutation loci specific enzyme cutting loci, and designing a dCAPS primer; c, predicating cleaved amplified polymorphism by combining genotype and phenotype data; d, verifying a dCAPS marker in a natural flooding tolerance extreme crowd; e, further verifying a WT-dCAPS1 marker in an F1 offspring flooding tolerance extreme crowd. The invention develops a dCAPS co-dominance marker co-separated from a flooding tolerance character of the chrysanthemum, the marker is named as the WT-dCAPS1, and the average verification accuracy is 78.9 percent in two crowds, so that the marker is preliminarily considered to be applied to chrysanthemum flooding tolerance molecular marker assistant breeding, beneficial to greatly shortening a breeding period, and has important theoretical and practical significance on cultivating new species of chrysanthemum with flooding tolerance.
