Embodiment 1

[0039] Example 1 Berberine promotes intraperitoneal injection of 3-NP-induced mitophagy in the brain of HD mice

[0040] Male Kunming mice, weighing 22-25 grams, were randomly divided into 4 groups, with 10 mice in the control group and 20 mice in the other groups. The four groups were control group, model group, low-dose berberine group (10mg / kg) and high-dose group (50mg / kg). The administration group was given intragastric administration for 10 consecutive days, once a day, and the control group and the model group were given the same volume of solvent (normal saline). On the 8th and 9th day of administration, the mice in the model group and the administration group were intraperitoneally injected with 3-NP, twice a day, with an interval of 12 hours each time, that is, 3-NP60mg / kg were injected at 8:00 and 20:00 on the 8th day , 8:00 and 20:00 on the ninth day, 3-NP 80 mg / kg was injected respectively, and the control group was intraperitoneally injected with the same volume...

Embodiment 2

[0055] Example 2 Berberine promotes mitophagy in the brain of HD rats induced by striatal injection of malonate

[0056] Male SD rats, weighing 200-220 grams, were randomly divided into 4 groups, 10 in each group. The four groups were control group, model group, low-dose berberine group (7.5mg / kg) and high-dose group (37.5mg / kg). Anesthetized by intraperitoneal injection of 3.5% chloral hydrate, the hair in the incision area on the top of the skull was cut off, disinfected with povidone iodine, and fixed on a brain stereotaxic apparatus. The left striatum (1.7mm in front of bregma, 2.7mm in the left side, and 4.8mm in depth) of rats in the model group and the treatment group was injected with 4 μl of malonate (6 μmol of malonate dissolved in 4 μl of normal saline) using a micro-injector. , the needle was retained for 5 minutes after injection; the control group was injected with the same volume of normal saline. After modeling, the administration group was given continuous i...

Abstract

The invention belongs to the technical field of medicine and relates to a medical application of BBR (berberine). The BBR can be used for promoting mitochondrion autophagy and improving nerve cell functions. The BBR can promote mitochondrion autophagy of nerve cells, increase expression of mitochondrion autophagy related genes, timely remove damaged mitochondria and improve the quality of the mitochondria. The effects of the BBR on intracerebral mitochondrion autophagy of HD (huntington's disease) model animals are observed through two HD animal models (a mouse intraperitoneal injection 3-NP model and a rat corpus striatum injection malonic acid model). Research results indicate that the BBR can improve motion and cognition disorders of the HD model animals by promoting mitochondrion autophagy of the nerve cells.

Description

technical field [0001] The invention relates to the field of medicinal chemistry, and relates to the medical use of berberine, in particular to the application of berberine in the preparation of drugs for promoting mitophagy, removing damaged nerve cell mitochondria, and improving nerve cell function. Background technique [0002] Huntington's disease (HD), also known as Huntington's disease or Huntington's disease, is an autosomal dominant neurodegenerative disease caused by cytosine in the IT-15 gene located in the 4p16.3 region of chromosome 4 - Caused by abnormal expansion of adenine-guanine (CAG) trinucleotide repeat sequence, when the number of CAG copies is greater than 36 and the disease occurs. Symptoms and signs of HD occur insidiously, with an average age of onset of 35-40 years old, adolescents (<20 years old) and old people (>70 years old) also have onset, and the survival period after onset is 15-20 years. Typical symptoms include chorea-like symptoms, c...
