[0023] The Bacillus genomic DNA extracted from the intestine of Bombina orientalis (Bacillussp.) captured from Maoershan, Heilongjiang Province, China was amplified by PCR using the Ness-5 / Ness-3 primer pair. The primer sequences are shown in Table 1. Referring to the N-terminal and C-terminal sequence homology of the Ness3A gene coding region published in GenBank, a pair of specific primers NESS5 / NESS3 were designed and synthesized, and NdeI and SalI restriction sites (indicated by arrows) were introduced at the 5' end, respectively.
