The extraction and enrichment technology comprises the following steps: taking a properamount of dried dactylicapnos root medicinal materials and crushing; putting into an extraction pot; adding a water solution which is 10 to 20 times of the weight of the dried dactylicapnos root medicinal materials; stirring and impregnating for one night; removing an impregnation solution on the next day; adding a 9 to 10 percent acid water solution which is 8 to 13 times of the weight of the dried dactylicapnos root medicinal materials; and carrying out hot refluxing and extracting for 2 to 5 times. The invention further discloses application of the protopine monomer in the dactylicapnos roots to cough and asthma relieving preparations, pain alleviating preparations, anti-arrhythmia preparations or anti-tumor preparations. The technology disclosed by the invention is suitable for extracting, enriching and purifying the protopine in the dactylicapnos roots and the purity of the protopine reaches 80 percent or more and can reach 95 percent or more after the protopine is further refined; and the whole technology has simple and feasible operation steps, high yield and low cost.
