Problems solved by technology

[0004] At this stage, all hospitals in various regions of my country are equipped with hematology analyzers for routine blood analysis of patients' blood. Among them, imported five-classification hematology analyzers are the main ones, but the original reagents for such hematology analyzers are expensive. And there are uncertainties in the quantity and cycle of supply, causing inconvenience to hospitals and patients

Embodiment 1

[0031] A white blood cell classification hemolysis agent for a five-classification blood cell analyzer, the formula is as follows:

[0032] Table 1

[0033] Brij93 1.6g / L DTAC 0.8g / L Potassium hydrogen phthalate 4.5g / L Succinic acid 0.4g / L sodium hydroxide 0.9g / L Sodium chloride 0.5g / L Isothiazolinone 0.2ml / L

[0034] Mix the above component ratios, add purified water to make up to 1L, stir at room temperature until completely dissolved, and filter with a 0.2 μm filter membrane, wherein the dyeing solution is 0.002% of polymethine dye, 3.1% of methanol and 96.8% of ethylene glycol %, the content is the mass percent concentration. Tested on Sysmex XN-1000, the blank value meets the requirements. Randomly take 8 fresh blood samples from people, and test them on Sysmex XN-1000 with original imported reagents and invented reagents respectively, and compare the experimental results. The experimental results are shown in T...

Abstract

The invention discloses a leukocyte classification kit for a five-classification hematology analyzer, and belongs to the technical field of cell detection. The kit is composed of a hemolytic agent anda staining solution. The hemolytic agent comprises a surfactant capable of rapidly dissolving erythrocytes and partially damaging cell membranes of leucocytes; cationic organic matters, which can becombined with anions in the leukocytes to make the leukocytes have morphological difference; and a buffer agent capable of adjusting the pH value of the hemolytic agent to 6.5-8.5. The staining solution comprises a polymethylene dye, methanol and ethylene glycol. The leukocyte classification kit for the five-classification hematology analyzer is easy to process and produce, accurate in detection result, stable in performance and lower in cost compared with an original imported reagent; the current situation that domestic imported instruments can only use expensive imported reagents is changed;and dependence on imported reagents is reduced.

Description

technical field [0001] The invention relates to a white blood cell classification kit for a five-classification blood cell analyzer, which belongs to the technical field of cell detection. Background technique [0002] Normal human white blood cells (WBC) are generally divided into five subgroups, namely lymphocytes (LYM), monocytes (MON), neutrophils (NEU), eosinophils (EOS) and basophils Granulocytes (BASO). These leukocyte subsets exist in certain proportions. But in disease, the number of certain blood cells increases or decreases. Therefore, in the field of clinical testing, the differential counting of white blood cells can be used for auxiliary diagnosis of some diseases. The detection of these five subgroups is detected through two channels on the five-differential blood cell analyzer, that is, one channel detects the number of the other four white blood cell subgroups, namely lymphocytes (LYM), monocytes (MON) , neutrophils (NEU), eosinophils (EOS), and another ...
