Embodiment 1

[0049] Example 1 Acute myocardial infarction significantly reduces the expression of BCAT1 and BCAT2

[0050] The mouse OGD-induced myocardial injury cell model and acute myocardial infarction model were constructed, and the expression differences of BCAT1 and BCAT2 in the two animal models and the control group were analyzed by Western blot.

[0051] Such as figure 1 As shown, the expressions of BCAT1 and BCAT2 were significantly reduced in the myocardial injury cell model induced by OGD, and the expressions of BCAT1 and BCAT2 were also significantly reduced in acute myocardial infarction model mice ( Figure 2-3 ).

Embodiment 2

[0052] Example 2 Overexpression of BCAT1 and BCAT2 can reduce myocardial damage caused by acute myocardial infarction

[0053] In order to explore the role of BCAT1 and BCAT2 in acute myocardial infarction, myocardial injection of adeno-associated virus carrying BCAT1 and BCAT2 genes was used to overexpress BCAT1 and BCAT2. After 4 weeks of transfection, 50% transfection rate was observed by immunohistochemical method, and then sham operation and acute myocardial infarction model were performed. The results showed that overexpression of BCAT1 and BCAT2 significantly reduced infarct size ( Figure 4 ), decreased the activity of CK and LDH ( Figure 5 ), and improved the histopathological characteristics of transfected mice ( Image 6 ). However, overexpression of BCAT1 and BCAT2 had no significant effect on sham-operated mice. In addition, overexpression of BCAT1 and BCAT2 had no effect on the expression of OXE-R. After acute myocardial infarction modeling, over-expression...

Abstract

The invention discloses application of branched aminotransferase 1 and / or branched aminotransferase 2. The invention for the first time finds that expression of branched aminotransferase 1 (BCAT1) andbranched aminotransferase 2 (BCAT2) in the cardiac tissue of an acute myocardial ischemia model mouse is abnormally reduced, and the phenomenon of abnormal reduction of expression also occurs in an oxygen-glucose deprivation induced myocardial injury cell model. Cardiac in-situ BCAT1 and BCAT2 protein overexpression is carried out on the mouse respectively, thus finding that myocardial ischemia injury can be effectively relieved. Therefore, the BCAT1 and the BCAT2 are proved to be able to serve as novel treatment targets for screening drugs for preventing, relieving and / or treating myocardialischemia injury.

Description

technical field [0001] The invention belongs to the field of medicine and relates to the application of branched chain aminotransferase 1 and / or branched chain aminotransferase 2. Background technique [0002] Acute myocardial infarction (AMI) is a very common disease worldwide with high mortality and morbidity, characterized by reduced oxygen and blood supply to the heart. At present, the diagnostic methods of myocardial infarction mainly include high-sensitivity troponin detection and electrocardiogram detection. High-sensitivity troponin detection has higher sensitivity but lower specificity for myocardial infarction. Therefore, ECG detection is still the main method for the diagnosis of myocardial infarction. The clinical treatment of acute myocardial infarction includes drug therapy, coronary artery stent implantation surgery, and coronary artery bypass surgery, combined with cardiac rehabilitation. The current commonly used therapeutic drugs mainly include aspirin, c...
