Abstract

The invention provides a one-step method rooting and transplanting technology for tissue culture seedlings of Fraxinus mandshurica. Semi-lignified adventitious individual plants obtained by tissue culture are cultured for 30 d in a strong seedling medium (WPM + 2.0 mg / L 6-BA + 20 g / L sucrose + 7 g / L agar), and then directly planted in a seedling culture bag containing a rooting medium (WPM + 1.4 mg / L IBA+0.7 mg / L NAA, without sucrose and agar) matrix (the ratio of turfy soil to vermiculite is 3:1), then the seedling culture bag is closed with a plastic pull bag, the plastic pull bag is opened7 d after culture under specific light, temperature and humidity conditions, the plastic pull bag is removed after 14 d, after 30 days of constant-temperature and constant-humidity culture, adventitious branches take root and grow out new leaves, and the survival rate is 63.33%. The technology simplifies rooting and transplanting into one step, is one of key technologies for rapid propagation of tissue culture seedlings of the Fraxinus mandshurica, saves the time and cost, and lays a solid foundation for the large-scale production of rapid propagation of the Fraxinus mandshurica.
