Abstract

The invention relates to a method for detecting klebsiella pneumoniae in a ready-to-eat fruit and vegetable agricultural product by mass spectrometry. The method comprises the following steps: S1, preparing klebsiella pneumoniae in ready-to-eat fruits and vegetables by specific selective culture; S2, establishing a method for separating klebsiella pneumoniae from fruits and vegetables; S3, extracting and separating suspected klebsiella pneumoniae protein components from fruits and vegetables; and S4, collecting a klebsiella pneumoniae protein spectrum by using matrix desorption ionization time-of-flight mass spectrometry, and performing identifying by adopting a characteristic peak fitting method. The core technology includes research and development of klebsiella pneumoniae selective medium components on the surfaces of ready-to-eat fruits and vegetables and establishment of a separation method, establishment of a pretreatment method of microbial mass spectrometry identification and establishment of microbial mass spectrometry conditions. According to the method for detecting and identifying the klebsiella pneumoniae in the ready-to-eat fruits and vegetables without standard agricultural products, the klebsiella pneumoniae in the ready-to-eat fruits and vegetables is detected and identified by adopting biochemical selective culture in combination with a mass spectrometry method, so that the klebsiella pneumoniae in the fruits and vegetables of agricultural products is accurately identified, more fruit and vegetable microorganism protein data information is obtained, and data is provided for traceability and pathopoiesis analysis of the klebsiella pneumoniae in fruits and vegetables.
