Problems solved by technology

[0004] In view of the above problems, the present invention provides a preparation method and application of a hydrogel with anisotropic stiffness gradient (gradient change of continuous stiffness value), which is used to at least partially solve the traditional method of using a uniform ECM stiffness hydrogel on bacteria. There are technical problems such as limitations in conducting research on infection

Abstract

The invention provides a preparation method and application of anisotropic stiffness gradient hydrogel. The preparation method comprises the steps of S1, preparing a lens groove-shaped polydimethylsiloxane mold; S2, adding a first preparation raw material into the lens groove of the polydimethylsiloxane mold and between the activated first cover glass, polymerizing and removing the mold to obtaina first layer of hydrogel in the shape of the lens groove; S3, adding a second preparation raw material between the first layer of hydrogel and hydrophobic second cover glass, and polymerizing to obtain a second layer of hydrogel; and S4, removing the second cover glass to obtain the anisotropic stiffness gradient hydrogel. The interface of the hydrogel provided by the invention is closer to a host tissue interface infected by bacteria, and the mechanical law of colonization and infection of bacteria on the gradient stiffness hydrogel interface can be more effectively disclosed through establishment of the physical characteristics.

Description

technical field [0001] The invention relates to the technical field of biomedical engineering, in particular to a preparation method and application of a hydrogel with anisotropic stiffness gradient. Background technique [0002] The colonization and infection of bacteria in the host are regulated by multiple factors, and the research field of their biological characteristics has been widely developed, but the research on the mechanical and physical characteristics still needs to be further explored. The host extracellular matrix (Extracellular matrix, ECM) is an important factor to ensure the integrity and mechanical homeostasis of the host tissue, and its matrix stiffness (Matrix stiffness) is the main mechanical property of the ECM. The polyacrylamide hydrogel technique for simulating different ECM matrix stiffnesses has been vigorously developed, and the matrix stiffness formed on the surface of each hydrogel was measured and prepared by Young's modulus method. The stif...
