Embodiment 1

[0027] Embodiment 1.DES-1 is solvent

[0028] Mix betaine (29.29g) with L-lactic acid (22.27g) and water (18.02g) at a molar ratio of 1:1:4, heat and stir at 50°C for 1h to form a clear and uniform natural deep eutectic solvent, Denote it as DES-1. Add 10mL of DES-1 into the Erlenmeyer flask, and then accurately weigh 0.5g Eucommia leaf powder into it, and extract it ultrasonically at 40°C for 30min with the ultrasonic power at 300W. After ultrasonic extraction, centrifuge at 4000r / min for 10min, and take the supernatant, which is the Eucommia extract. After the supernatant is diluted 25 times with an aqueous solution, it is detected by high performance liquid chromatography (HPLC). According to the standard curve of the standard, the active ingredient yield in the Eucommia extract is calculated, and the active ingredient yield=biologically active ingredient content / weight of Eucommia raw material mg / g. See Table 1 for the results.

Embodiment 2

[0036] Embodiment 2.DES-2 is solvent

[0037] In Example 1, DES-1 was changed to choline chloride (34.95g), mixed with urea (15.01g) and water (18.02g) in a molar ratio of 1:1:4, and heated and stirred at 50°C for 1h to form A clear and uniform natural deep eutectic solvent, denoted as DES-2. Other operations were the same as in Example 1, and the yields of aucubin, geniposidic acid, chlorogenic acid, and rutin in the Eucommia extract were measured in Table 2.

[0038] Table 2 Comparison of DES-2 and water extraction results

[0039]

[0040] It can be seen that the yields of Eucommia ulmoides extracted with natural deep eutectic solvent: aucubin, geniposidic acid, chlorogenic acid, and rutin are all higher than water extracts.

Embodiment 3

[0041] Embodiment 3.DES-3 is solvent

[0042]Change DES-1 in Example 1 to choline chloride (34.95g) and L-lactic acid (22.27g), water (18.02g) at a molar ratio of 1:1:4, heat and stir at 50°C for 1h to form a clear Uniform natural deep eutectic solvent, denoted as DES-3. Other operations were the same as in Example 1. The measured yields of aucubin, geniposidic acid, chlorogenic acid and rutin in the Eucommia ulmoides extract are shown in Table 3.

[0043] Table 3 Comparison of DES-3 and water extraction results

[0044]

[0045] It can be seen that the yield of chlorogenic acid is 30.15 mg / g, which is 11.17% higher than that of the water extract in Comparative Example 1; the yield of rutin is 3.22 mg / g, which is 110.46% higher than that of the water extract in Comparative Example 1.

Abstract

The invention discloses a method for preparing eucommia ulmoides extract by using a natural eutectic solvent. The method comprises the following steps that: eucommia ulmoides powder is added into the natural eutectic solvent according to a material-liquid ratio of (1: 1)-(1: 100g/mL), and ultrasonic-assisted extraction is performed for 10-90 minutes at 20-80 DEG C and 50-500W, wherein the natural eutectic solvent is composed of a hydrogen bond donor, a hydrogen bond acceptor and distilled water according to the mass ratio of 1: (1-30): (1-50), wherein the hydrogen bond donor comprises L-lactic acid, sorbitol, urea, DL-malic acid, D-fructose, glucose and citric acid, and the hydrogen bond acceptor comprises choline chloride, betaine and citric acid; centrifugation is performed, supernatant is taken, and the supernatant is further concentrated and dried, so that the eucommia ulmoides extract is obtained. The method is safe, economical and recoverable; various active ingredients such as aucubin, geniposidic acid, chlorogenic acid and rutin in eucommia ulmoides can be extracted; and compared with that of a water extract, the content of the active ingredients in the natural eutectic solvent extract is remarkably increased, the extraction efficiency is high, and the components of the natural eutectic solvent can also be used as functional components in functional food and cosmetics; and the step of removing an extraction solvent in a traditional process is omitted. The method is more economical and efficient.

Description

Technical field: [0001] The invention relates to a method for preparing Eucommia ulmoides extract by utilizing a natural deep eutectic solvent. Background technique: [0002] Eucommia ulmoides Oliv., also known as Sizhong, Sixian, etc., is a perennial deciduous tree belonging to the genus Eucommia in the family Eucommia, and is a unique valuable economic tree species in my country. Eucommia is widely planted in my country, mainly distributed in Hunan, Guizhou, Sichuan, Guangxi, Henan, Shaanxi and other places. The "Shen Nong's Materia Medica" of the Han Dynasty in my country recorded the efficacy of Eucommia ulmoides, and listed Eucommia as the top grade. Li Shizhen of the Ming Dynasty wrote a great book "Compendium of Materia Medica" in 1578, which recorded the origin and efficacy of Eucommia in more detail. In recent years, scholars at home and abroad have made in-depth research on the metabolites, pharmacological activities and clinical applications of Eucommia ulmoides...
