Problems solved by technology

Although many studies have shown that PHLPP2 can inhibit the occurrence and development of tumors, whether it plays a role in vitamin C inhibiting the occurrence and development of pancreatic cancer and whether it is a potential biomarker for vitamin C in the treatment of pancreatic cancer has not yet been clarified

Embodiment 1

[0046] In this example, the experiments and analysis of vitamin C's inhibition of pancreatic cancer cell proliferation, cell migration, and promotion of pancreatic cancer cell apoptosis, the effect of PHLPP2 on the RAS / RAF signaling pathway, and the relationship between PHLPP2 expression and prognosis were carried out.

[0047] Cell culture and reagents: Human pancreatic cancer cells Capan-1 and PANC-1 were purchased from the Cell Bank of the Chinese Academy of Sciences. Cells were cultured in DMEM+10% FBS. Vitamin C was purchased from Sigma, dissolved in 1×PBS;

[0048] Quantitative PCR to detect the level of PHLPP2 mRNA: Total RNA was extracted by Trizol method. RNA was reverse transcribed into cDNA using Hifair II 1st Strand cDNA Synthesis SuperMix kit purchased from Yisheng Company. The cDNA was amplified by PCR using the SYBR Green Mix kit purchased from Yisheng Company, and β-actin was used as an internal reference, and the gene transcription level was analyzed by ΔΔCT...

Abstract

The invention belongs to the technical field of biology, and relates to application of phosphatase PHLPP2 as a biomarker, in particular to an application of PHLPP2 in preparation of a prognostic marker preparation for treating pancreatic cancer through vitamin C. The PH structural domain of the phosphatase is rich in leucine repeat protein phosphatase 2, and corresponding conserved sites of Akt, RAF1 and the like can be directly dephosphorylated, so that cell apoptosis is promoted, and cell proliferation and migration are inhibited; the tests show that the expression level of the PHLPP2 is reduced, the killing effect of the vitamin C on pancreatic cancer tumors can be inhibited, the high expression of the PHLPP2 is positively related to the lifetime of a patient and is negatively related to TNM staging, and the PHLPP2 can be used as a potential biomarker of the vitamin C for treating the pancreatic cancer and is further used for preparing a prognosis marker preparation of the vitamin C for treating the pancreatic cancer.

Description

technical field [0001] The field of biotechnology of the present invention relates to the use of a phosphatase PHLPP2 as a biomarker, in particular to the application of PHLPP2 in the preparation of vitamin C for the treatment of pancreatic cancer prognosis marker preparations. Background technique [0002] Pancreatic cancer is reported to be a highly lethal disease, with the latest data showing approximately 56,770 new cases and 45,750 deaths annually in the United States. Clinical practice shows that due to the lag of diagnosis and drug resistance of treatment, the prognosis of pancreatic cancer patients is extremely poor, and the 5-year survival rate remains at 6%. Studies have reported that vitamin C has tumor-killing effects both in vitro and in vivo, but recent clinical trials and case studies of pancreatic cancer have failed to clarify the clinical efficacy of vitamin C or only explain that vitamin C can improve the quality of life of patients and enhance chemotherapy...
