[0054] Construction of prognostic risk prediction model for cervical cancer

[0055] 1. Data download and arrangement

[0056] The gene expression data of cervical cancer samples and normal samples, as well as the clinical information data corresponding to cervical cancer patients, were downloaded from the TCGA database, including 306 cancer tissue samples and 3 normal tissue samples as a training set.

[0057] NRGs (hsa04217) were downloaded from KEGG (Kyoto Encyclopedia of Genes and Genomes), and combined with literature reports, 211 NRGs were finally included for subsequent analysis. Use the perl script to extract the expression data of NRGs from the training set expression profile data, and normalize the gene expression data to obtain the NRGs expression matrix; use the R package limma to analyze the differential expression of NRGs in the training set, with P2 was used as the cutoff value for screening differential genes, and finally a total of 17 differentially expressed ...

Abstract

The invention discloses a molecular marker combination for predicting prognosis conditions of cervical cancer patients and application of the molecular marker combination. According to the invention, three screened NRGs including CHMP4C, SLC25A5 and VDAC1 are taken as a molecular marker combination, a risk prediction model with three NRGs is established, and cervical cancer patients are divided into a high risk group and a low risk group. According to the risk prediction model, the AUC value corresponding to the one-year lifetime prediction is 0.694, the AUC value corresponding to the three-year lifetime prediction is 0.703, the AUC value corresponding to the five-year lifetime prediction is 0.664, and better accuracy and specificity are achieved. And the prediction performance of the risk prediction model in the invention is verified in a verification set. The model provided by the invention can effectively predict prognosis of a cervical cancer patient, which is beneficial to selection of a clinical treatment scheme and further realizes early intervention and early treatment of cervical cancer.

Description

technical field [0001] The invention belongs to the field of medical detection, and particularly relates to a molecular marker combination for predicting the prognosis of cervical cancer patients and its application. Background technique [0002] Cervical cancer is a common gynecological disease and one of the leading causes of cancer deaths in women. At present, the main treatment method for cervical cancer is radical hysterectomy combined with radiotherapy and cisplatin chemotherapy, but some patients will still relapse after surgery or radiotherapy, and the prognosis of relapsed patients is poor. In order to further improve the therapeutic effect of cervical cancer and formulate precise treatment strategies, oncologists need to predict the prognosis of cervical cancer patients. [0003] Currently, the FIGO (Federation International of Gynecology and Obstetrics) staging system is commonly used in clinical practice to predict the prognosis of cervical cancer patients. For...
