[0165] instance number 1

[0166] The following examples were conducted to test the ability of filter devices made in accordance with the present disclosure to draw fluid culture medium from a stirred tank bioreactor without simultaneously drawing out or damaging the biological cells contained in the bioreactor.

[0167] The filter equipment used in the following experiments was similar to Figure 4A Design shown in . The length of the filter member is approximately 1.5in 2 , and the surface area of the outer surface of the filter member is 0.65 in. 2 . The filter members are made of sintered stainless steel fibers and have an absolute pore size of 3-4 microns. The inner diameter of the tubular member attached to the filter member was 6.35 mm.

[0168] T cells at 3-6.5 x 10 6 Cells / mL were seeded into 1-liter bioreactors. The bioreactor is a stirred tank bioreactor. The cell culture was maintained in the bioreactor for 1 day, during which time the fluid medium was ...

Abstract

A filter apparatus is disclosed for withdrawing a fluid medium from a bioreactor during growth of a cell culture within the bioreactor. A method for culturing cells in a bioreactor is also disclosed. The filter apparatus includes a hollow tubular member attached to a filter member. The filter member has an aperture and volume capable of withdrawing fluid medium from the bioreactor at relatively high flow rates. Without withdrawing biological cells from the bioreactor and without damaging or damaging the cells. The filter apparatus of the present disclosure allows many process improvements.

Description

Background technique [0001] Bioreactors are widely used within the biopharmaceutical industry, which are devices that can carry out biological reactions or processes on a laboratory or industrial scale. Bioreactors can be used in batch applications where the biological material supplied to the bioreactor remains in the bioreactor until the end of the reaction time. Alternatively, bioreactors can be used in perfusion applications where the fluid medium contained within the bioreactor is periodically or continuously withdrawn and resupplied to the bioreactor to supplement the fluid medium contained in the bioreactor nutrients and for the possible removal of harmful by-products produced during the process. [0002] Bioreactors are used, for example, for the production of biological agents, which are biopharmaceuticals produced from living organisms. Bioreactors are also used in immunotherapy, a type of treatment that boosts a patient's immune system to fight cancer, infection, ...
