Embodiment 1

[0052] Citric acid biomass was pretreated with 4% aqueous sodium hydroxide (NaOH) by autoclaving at 120° C. for 1 hour, a step that removed excess protein and other undesirable materials. The biomass was then washed thoroughly with deionized water until the pH was approximately 7.0. The washed material is mixed with concentrated hydrochloric acid (HCl) and water to form a mixture containing 10-15% hydrochloric acid and 5-6% biomass (by dry weight of biomass). The mixture was heated to reflux. Samples were taken from time to time and the reaction was analyzed by Dionex HPLC high pressure liquid chromatography (trade name DX-500).

[0053] The results are shown in Figure 1, which is an indicative graph of glucosamine production and shows that glucosamine production increased continuously within the 8 hour reaction, but the amount of glucose decreased after 4 hours. The yield of glucosamine after 8 hours was 14%. The chromatogram of this product is shown in Figure 2.

Embodiment 2

[0056] Example 1 was repeated, but the pretreated biomass was maintained at reflux for 13 hours. The glucosamine produced has a purity of over 98%.

Abstract

Glucosamine suitable for human or animal consumption is disclosed. The glucosamine is derived from microbial biomass containing chitin. Suitable starting materials include substantially uniform microbial fungal sources, such as fungal sources derived from Aspergillus sp., Penicillium sp., Mucor sp. and combinations thereof. Methods of producing glucosamine by acid hydrolysis of fermented fungal biomass are also disclosed.

Description

[0001] This application is a PCT application filed on February 15, 2002 by CARGILL Corporation, a US national and resident corporation, designating all countries except the United States. field of invention [0002] The present invention relates to glucosamine compositions and methods of making glucosamine compositions. Background of the invention [0003] Glucosamine is a nutraceutical supplement that has been shown to significantly reduce arthritis and joint pain therapeutically. Although the mechanism is not fully understood, it is believed that glucosamine functions to help restore cartilage to reduce inflammation in the joints, thus providing significant benefit to the patient. [0004] At present, glucosamine is basically derived from obtained natural raw materials, such as crustaceans and other aquatic organisms. The shell and exoskeleton components of these organisms are converted into glucosamine using different production te...
