[0018] (1) Preparation of reference substance solution: take an appropriate amount of hydroxysafflor yellow A HSYA reference substance, add distilled water to make a solution containing 0.15mg in 1ml.
[0019] (2) Preparation of the test solution: take 15 mg of the sample, add water and dilute it to a 25 ml volumetric flask.
[0020] Accurately draw 20ul each of the above-mentioned reference substance solution and the test solution respectively, inject it into a liquid chromatograph, measure it, and obtain it.
Embodiment 1
[0021] Embodiment one safflower yellow pigment extraction process
[0022] Add 500g of safflower, add deionized water at 50°C and soak for 3 times, the amount of water added is 16 times, 10 times, 10 times of the weight of safflower, respectively, and the extraction time is 2 hours, 2 hours, 1 hour respectively. The three extracts were combined, adjusted to pH=2.0, and filtered. The filtrate was passed through macroporous resin D-4020, washed with water for 3 times the column volume, and discarded. The resin column continued to be eluted with water, and 6 times the column volume of water was collected. The eluate was concentrated through a reverse osmosis membrane to remove 80% of water, and the concentrate was dried under reduced pressure to obtain safflower yellow with a yield of 1.98% and a content of hydroxy safflower yellow of 32.0%.
Embodiment 2
[0023] Embodiment two safflower yellow pigment extraction process
[0024] For 500g of safflower, add deionized water and soak for 3 times at 60°C. The amount of water added is 16 times, 10 times, and 10 times of the weight of safflower, and the extraction time is 2 hours, 2 hours, and 1 hour respectively. The three extracts were combined, adjusted to pH=2.5, and filtered. The filtrate was passed through macroporous resin D-4020, washed with water for 3 times the column volume, and discarded. The resin column continued to be eluted with water, and 6 times the column volume of water was collected. The eluate was concentrated through a reverse osmosis membrane to remove 80% of water, and the concentrate was dried under reduced pressure to obtain safflower yellow with a yield of 1.87% and a content of hydroxy safflower yellow of 31.5%.
