Embodiment 1

[0049] The preparation of embodiment 1.0.6EFS freezing liquid:

[0050] Put 0.6g of polysucrose in a 10ml volumetric flask, add PBS to near the mark, and let it stand at 4°C to dissolve; after it is completely dissolved, add 0.3424g of sucrose, and let it stand at 4°C to dissolve; after it is completely dissolved, add 0.021g of BSA (do not Shake to prevent foaming), stand at 4°C to dissolve; after complete dissolution, add PBS to make up to the mark; after mixing the liquid in the volumetric flask, take 9.2ml of the solution in the volumetric flask and 0.8ml of pure ethylene glycol, seal , put it in the refrigerator at 4°C, and set aside.

Embodiment 2

[0051] The preparation of embodiment 2.0.1EFS freezing liquid:

[0052] The preparation steps are the same as in Example 1, 0.1 g of polysucrose is added, and the amounts of sucrose, pure ethylene glycol and bovine serum albumin are the same as above.

Embodiment 3

[0053] The preparation of embodiment 3.0.2EFS freezing liquid:

[0054] The preparation steps are the same as in Example 1, adding 0.2 g of polysucrose, and the amounts of sucrose, pure ethylene glycol and bovine serum albumin are the same as above.

Abstract

The invention discloses a freezing liquid for preserving embryo, especially the freezing liquid for preserving sampled embryo. The freezing liquid comprises ethylene glycol and sucrose, and is characterized by comprising 9-65g of polysucrose, 75-85mL of ethylene glycol, 30-40g of sucrose, and 910-930mL of PBS buffer solution containing 0.0021 g/mL bovine serum albumin (BSA). A preparation method comprises the steps of dissolving ethylene glycol, sucrose and polysucrose in the BSA-containing PBS buffer solution. The freezing liquid fully utilizes synergic effects of the four components that ethylene glycol, sucrose, arabinogalactan and bovine serum albumin are coordinated and supported one another, and has higher development rate of thawed sampled embryo in comparison with conventional ethylene glycol freezing liquid.

Description

technical field [0001] The invention relates to a freezing liquid for preserving embryos, in particular to a freezing liquid for preserving sampled embryos. The invention also relates to the preparation method and application of the freezing liquid. Background technique [0002] Embryo freezing is a method of embryo preservation. Embryos are placed in freezing liquid for protection and special cooling procedures are adopted to stop the metabolism of embryos at ultra-low temperatures (liquid nitrogen -196°C, liquid helium -269°C). When the embryos need to be used, they can be restored to viability through specific thawing procedures, so as to achieve the purpose of long-term preservation of embryos. [0003] For gender identification and other genetic testing and analysis, it is often necessary to use micromanipulation techniques to cut off a small part of embryonic tissue for analysis, and freeze the remaining part for future use. [0004] However, after the embryo is cut ...
