Problems solved by technology

At present, there are reports on taro tissue culture research at home and abroad, but most of them focus on the induction of callus at the shoot tip, which is not conducive to maintaining the stability of the variety.

Embodiment Construction

[0024] The present invention will be further described below by obtaining test-tube plantlets under the condition of ex-vivo based on the stem tip of Jingjiang sweet taro, a geographical landmark agricultural product in China.

[0025] 1 Materials and methods

[0026] 1.1 Test material

[0027] Potted taro is adopted, and the stem tip is selected as the explant.

[0028] 1.2 Method

[0029] 1.2.1 Handling of sterile materials

[0030] Choose well-preserved taro with no rot spots, and pass through 0.2% K 2 MnO 4 After sterilizing for 5 minutes, arrange them neatly in the turnover box, cover 2 / 3 of the whole taro block with sand, and place it in the light incubator to wait for it to germinate. When the bud grows to 1-3cm, cut off the stem tip, peel off the outer 2-3 layers of leaves, rinse with running water for about 10 minutes, soak in 75% alcohol for 30s on the ultra-clean table, and treat with two kinds of disinfectants for 3 time periods respectively For sterilization...

Abstract

The invention discloses a rapid propagation method for performing tissue culture by using a taro stem tip, belonging to the technical field of biology. The method comprises the following steps of: selecting an aseptic taro stem tip as an explant; and performing a series of operating steps such as bud induction, subculture multiplication, rooting culture and the like to form a complete test-tube plantlet. The method comprises treatment methods for culture medium components at each abovementioned stage and induction culture. A seedling with two or more leaves can grow by performing primary culture on the taro stem tip for about 20 days, so that the process of primary culture is accelerated greatly; the growth coefficient is 4.3 on average after one-time secondary culture, secondary culture can be performed for five times generally to culture 1,470 (4.35) seedlings, and two batches can be cultured every year to form 2,940 tissue culture seed tuber seedlings, so that large-scale commercial production of taros is facilitated; and the method is also easy for obtaining regeneration plants with high genetic stability and low aberration rates.

Description

technical field [0001] The "rapid propagation method for tissue culture by using taro stem tips" of the present invention is to obtain test-tube plantlets by tissue culture of taro stem tips in vitro, and belongs to the field of biotechnology. Background technique [0002] Jiangsu Province has many varieties of local characteristic taro, which has good nutritional and health care quality, and contains a variety of vitamins. It can be used as food and vegetables. It is a tonic suitable for all ages. It has broad processing prospects and is famous and sold well Domestic and foreign markets. At present, the production of local characteristic taro in Jiangsu Province usually adopts the method of self-reserved seeds for breeding, and the problem of species degradation is more prominent, and the disease resistance and stress resistance are reduced. Moreover, there are very few domestic studies on its tissue culture and rapid propagation technology. Therefore, the research on the ...
