specific Embodiment approach 1

[0016] Specific embodiment one: In this embodiment, the method of using nematode larvae to detect plasticizers is as follows:

[0017] 1. Weigh 1.0 g of cholesterol, dissolve it in absolute ethanol, and prepare a cholesterol ethanol solution with a concentration of 10 mg / ml;

[0018] 2. Preparation of phosphate buffer: weigh 13.6g KH 2 PO 4 , 1.8g KOH, add distilled water to 100mL, adjust the pH value to 6.0;

[0019] 3. Mix 15-20g agar powder, 2-3g peptone, 0.10-0.12g CaCl 2 , 0.11~0.13g MgSO 4 Add 2~3g NaCl and 2~3g NaCl into a 1000mL beaker, add 1ml cholesterol ethanol solution and 25mL phosphate buffer, and distill the volume to 1000ml with distilled water;

[0020] 4. Seal the beaker, put it in a high-pressure steam sterilizer at 120°C for 15-25 minutes, then cool it down to 50-60°C, add 0.5-5ml of white wine to be tested under a sterile environment, and then put the mixture in a sterile Pour it into a petri dish under the environment, and dry it in a sterile environ...

specific Embodiment approach 2

[0022] Specific embodiment two: the difference between this embodiment and specific embodiment one is that in step two, 16~19g agar powder, 2g peptone, 0.10g CaCl 2 , 0.11g MgSO 4 and 2g NaCl were added to a 1000mL beaker. Others are the same as in the first embodiment.

specific Embodiment approach 3

[0023] Specific embodiment three: what this embodiment is different from specific embodiment one is that in step 2, 15g agar powder, 2g peptone, 0.10g CaCl 2 , 0.11g MgSO 4 and 2g NaCl were added to a 1000mL beaker. Others are the same as in the first embodiment.

Abstract

The invention discloses a method for detecting a plasticizer by nematode larvae, and relates to a method for identifying the plasticizer. The method solves the problems in a conventional plasticizer detection method that the operation is complex, the dependence degree for a device is large, and the cost is high. The method comprises the steps as follows: step one, preparing a cholesterol ethanol solution; step two, preparing a phosphate buffer solution; step three, preparing nematode culture media; and step four, coating the nematode culture media with a colon bacillus OP50 bacterium solution uniformly, culturing the culture media for 8-12h at the temperature of 37 DEG C, putting in 8-12 nematode larvae in the L2 period, culturing for 12h, and then observing the death rate of nematodes in the L2 period. If the death rate of the nematode larvae is higher than 10%, the fact that a distilled spirit to be detected contains the plasticizer is indicated. The detection method has low requirements for experimental conditions, and does not require any special high-precision analytical instruments or detection devices, the experiment can be operated conveniently, the cost is low, and the method can be mastered easily.

Description

technical field [0001] The invention relates to a method for detecting plasticizers. Background technique [0002] Plasticizers are phthalate homologues. Including DEHP, DINP, DNOP, DBP, DMP, DEP and other 6 kinds of phthalate plasticizers. On June 1, 2011, the Ministry of Health issued an emergency announcement that phthalates (also called phthalates) ) substances, included in the list of non-edible substances that may be illegally added to food and food additives that are easily abused. However, at the end of 2012, it was exposed that the plasticizer of "drunkard" wine exceeded the standard by 2.6 times, which caused people's panic and doubts about the plasticizer and the food safety of liquor, especially high-end wine. [0003] As an environmental hormone, plasticizers can cause immunity and reproductive decline in humans and animals. At present, the tolerable daily intake range of 60 kg adults in various countries is 1.2-8.4 mg. Some studies have shown that the potent...
