Embodiment 1
[0031] Embodiment 1, the preparation of Auricularia officinalis protein extract
[0032] 1. Prepare the fruiting body of Auricularia auricula
[0033] Inoculate Auricularia polytricha (Mont.) Sacc. No. 3 slant strains into the first-level medium for activation, culture at a constant temperature of 25°C, and inoculate it into the second-level medium after the mycelium is full of the test tube in a constant temperature culture room at 25°C until the hyphae are overgrown, inoculate the second-grade species into a cultivation bag with a cultivation medium for growth at 25°C, and scratch the bacteria after the strain is full of the cultivation bag and transplant it into the In the greenhouse, the fruiting conditions should keep the humidity above 90% and the temperature at 20-25°C. Collect the first tide fruiting bodies to obtain No. 3 fruiting bodies of Auricularia polytricha (Mont.) Sacc.

Embodiment 2
[0042] Example 2, Auricularia auricularia protein extract inhibits tumor cell proliferation experiment
[0043]1.1 Tested cell lines
[0044] Human liver cancer HepG2 cells (purchased from ATCC, USA), human breast cancer MCF-7 cells (purchased from ATCC, USA) and human lung adenocarcinoma A-549 cells (purchased from ATCC, USA).
[0045] 1.2 Experimental method
[0046] 1.2.1 Cell lines and cell culture
[0047] Human liver cancer HepG2 cells, human breast cancer MCF-7 cells and human lung adenocarcinoma A-549 cells were activated and passaged according to conventional culture methods. Among them, the subculture and activation medium of human liver cancer HepG2 cells is DMEM-high glucose + 1% double antibody + 10% FBS (in DMEM-high glucose (Hyclone, SH30022.01B) a mixture of penicillin and streptomycin (penicillin 10000U / mL, streptomycin 10000μg / mL) and fetal bovine serum (FBS, Hyclone, SV30087.02), so that the final volume percentage of the former two is 1%, and the volume ...

Abstract
The invention discloses an auricularia polytricha protein extract and anti-tumor application thereof. The application is application of the auricularia polytricha protein extract in preparation of a product for restricting tumor cell proliferation. The auricularia polytricha protein extract is prepared according to the method comprising the following steps of: 1) extracting by water after crushing auricularia polytricha sporocarp, collecting a water-soluble matter to obtain an auricularia polytricha water-soluble extract; 2) carrying out ammonium sulfate precipitation of which the saturation level is 80% on the auricularia polytricha water-soluble extract, collecting sediments, and obtaining the auricularia polytricha protein extract after dialyzing the sediments by deionized water. Multiplication of HepG2, MCF-7 and A-549 cells is obviously restrained after the human liver cancer HepG2 cell, the human breast cancer MCF-7 cell and the pulmonary adenocarcinoma A-549 cell are processed by the auricularia polytricha protein extract for 72 hours; the IC50 values are 45 mug / mL, 20 mug / mL, and 25 mug / mL respectively.
Description
technical field [0001] The invention relates to the protein extract of the fungus and its anti-tumor application. Background technique [0002] Auricularia is a fungus of A. polytricha in the order Auriculares, family Auriculariaceae, and the genus Auricularia. It is an important edible fungus and also used as medicine. Rich in protein, various mineral elements and vitamins, the fungus polysaccharide rich in the villi layer on the back of the fruiting body has high anti-tumor activity. In the current research, there are many researches on the cultivation technology of Auricularia edulis, the extraction technology of Auricularia edulis polysaccharide and the function of polysaccharide. Zheng Lixue et al. conducted a more detailed study on the submerged fermentation process of Auricularia auricola mycelia, and optimized the medium and process conditions of Auricularia auricola submerged fermentation by taking the dry mass concentration of mycelium as an investigation index. ...
