Abstract

The method comprises the following steps: (1) preparation of culture media: including the following ingredients and contents in each tissue culture stage: 1) bud inducing culture media: MS, and 1.0-5.0 mg/L of 6-BA and 0.1-0.5 mg/L of NAA; 2) adventitious bud proliferation culture media: MS, and 1.0-3.0 mg/L of 6-BA and 0.1-0.3 mg/L of NAA; 3) strong seedling culture media: MS, and 0.5-2.0 mg/L of 6-BA and 0.1-0.2 mg/L of NAA; 4), rooting culture media: MS and 0.05-0.2 mg/L of NAA; controlling the ph of the culture media under each condition to 5.8, adding 30 g/L of glucose and 6g/L of agar, and culturing at a temperature of 26 DEG C or 24 DEG C under the solar irradiation of about 80 micromol/ms.
