Embodiment 1

[0015] Embodiment 1 Rutin laboratory preparation 1

[0016] Sophora japonica is crushed and sieved to 60 mesh. Weigh 100g of crushed Sophora japonica, put it into a 1000ml three-neck bottle with stirring, then add 50% dilute ethanol in a volume 5 times the weight of Sophora japonica, soak for 1 hour, stir for 5 minutes every 30 minutes, and continue for 3-5 hours. This procedure was repeated 3 times. Remove the Sophora japonica residue by suction and filtration, combine the extracts, concentrate under reduced pressure to recover most of the ethanol, and transfer it to a crystallization flask while it is hot. Cool and stir for about 1-2 hours to crystallize and precipitate crude rutin, filter, and dry to obtain 23.8 g of crude rutin for later use. Take 20g of the crude rutin from the previous step, stir and dissolve it with 100ml of 0.5% sodium hydroxide aqueous solution, pass it through a fine filter, then cool and stir, adjust the pH to 6-7 with concentrated hydrochloric ac...

[0017] Embodiment 2 Rutin laboratory preparation 2

[0018] Sophora japonica is crushed and sieved to 40 mesh. Weigh 100g of crushed Sophora japonica, put it into a 1000ml three-neck bottle with stirring, then add 60% dilute ethanol with a volume 5 times the weight of Sophora japonica, soak for 1 hour, stir for 5 minutes every 30 minutes, and continue for 3-5 hours. This procedure was repeated 3 times. Remove the Sophora japonica residue by suction and filtration, combine the extracts, concentrate under reduced pressure to recover most of the ethanol, and transfer it to a crystallization flask while it is hot. Cool and stir for about 1-2 hours to crystallize, and the crude rutin is precipitated, filtered, and dried to obtain 23.5 g of the crude rutin for later use. Take 20g of the crude rutin from the previous step, stir and dissolve it with 100ml of 0.5% sodium hydroxide aqueous solution, pass it through a fine filter, then cool and stir, adjust the pH to 6-7 with concentra...

Embodiment 3

[0019] Embodiment 3 industrialized preparation of rutin

[0020] Sophora japonica is crushed and sieved to 60 mesh. Weigh 100kg of crushed Sophora japonica, add it to a 1000L extraction tank with stirring, then add 45% dilute ethanol 5 times the volume of Sophora japonica, soak for 1 hour, stir for 5 minutes every 30 minutes, and continue for 3-5 hours. This procedure was repeated 3 times. Use an automatic discharge centrifuge to separate the Sophora japonica residue and the extract, and combine the extract to the storage tank. Transfer the dilute ethanol extract of Sophora japonica obtained in the previous step into a concentration pot, concentrate under reduced pressure to recover most of the ethanol, and transfer it to a crystallization tank while it is hot. Cool and stir for about 1-2 hours to crystallize, and the crude rutin is precipitated, filtered, and dried to obtain 24.1 kg of crude rutin for later use. Take 20kg of crude rutin from the previous step, stir and dis...

Abstract

The invention provides a green extraction process of natural active flavone-rutin. The process is to extract, separate and purify high-purity and high-content rutin from sophora flower bud. The process comprises the following steps of: crushing dried sophora flower bud into 40 to 60 meshes; transferring into an extracting tank; adding 5 to 8 times dilute alcohol for soaking and extracting; separating the extracting liquid and residue to obtain the filtrate; treating the filtrate by pressure-reduced concentration to remove most of alcohol; transferring into a crystallizing tank; devitrifying to obtain crude rutin; dissolving the crude rutin with 0.5% of strong base aqueous solution; processing through a fine filter; regulating the pH to be 6 to 7; crystallizing; filtering; and drying to obtain fine rutin. The method has the advantages that the production process is green and environmentally friendly, the equipment is simple, and thus industrial popularization is facilitated.

Description

technical field [0001] The invention relates to an extraction and separation process of natural active active flavonoids, in particular to high-efficiency extraction with ethanol solution as an extraction solvent, separation and concentration to obtain crude rutin, which is then refined with pure water, alkali and acid, filtered and dried to obtain High purity and high content refined rutin. Background technique [0002] Rutin, also known as rutin, rutin, Luotong or quercetin-3-O-rutinoside, also known as vitamin P. Its chemical structure is a glycoside formed between the flavonol quercetin and the disaccharide rutinobiose. As the most widely active flavonoid glycoside, it can lower blood pressure, cool blood and activate blood, and prevent hemorrhagic complications caused by high blood pressure and diabetes. Food additives are used as antioxidants and pigments. Its derivative, troxerutin, inhibits the aggregation of platelets and has the effect of preventing thrombosis. ...
