Problems solved by technology

However, due to the inability to accurately distinguish the released individuals from the wild individuals and their quantity in the recaptured population, it is impossible to scientifically assess the supplementary level of the released population to wild resources, analyze the dynamic changes of wild resources and formulate a scientific release plan. This is an urgent release of Penaeus japonicus major issues to be addressed

Embodiment 1

[0025] DNA extraction and PCR from Penaeus japonicus:

[0026] (1) DNA extraction: Trizol method extraction.

[0027] A. Take 1000μL Tris-cl into a 2ml centrifuge tube;

[0028] B. Take an appropriate amount of tissue (about 2-3mg), and fully cut it with sterilized scissors;

[0029] C. After soaking for 45min, 4℃, 12000rpm, 10mim;

[0030] D. Discard the supernatant, add 600 μL Tris-buffer, 50 μL 10% SDS, 7 μL proteinase K, water bath at 55°C for 2 h, until complete digestion and the solution is clear;

[0031] E. Add 350 μL phenol, 350 μL chloroform:isoamyl alcohol (24:1), shake until completely mixed; 4°C, 12000rpm, 10mim;

[0032] F. Take the supernatant into a new 2ml centrifuge tube, add the same volume of chloroform:isoamyl alcohol (24:1) as the supernatant, shake until completely mixed; 4°C, 12000rpm, 10mim;

[0033] G. Take the supernatant into a new 1.5ml centrifuge tube, add 2 times the volume of supernatant with ice anhydrous ethanol, mix well, let stand at -20...

Abstract

The invention relates to an evaluation method for the releasing effect of Marsupenaeus Japonicus. The method includes: conducting amplification and sequencing on maternally inherited mitochondrial sequences, analyzing a genetic distance by relevant software, and carrying out system construction, haplotype analysis and the like to analyze sequence differences and haplotype distribution of wild populations and breeding populations, counting the intra-population frequency of each haplotype, selecting one or several specific parent shrimps with specific molecular markers to breed first filial generation of small shrimps for releasing, thus determining the proportion of individuals in recaptured populations, and performing comparison with the proportion of the molecular markers before releasing so as to determine the releasing effect of the batch. The method provided by the invention has the characteristics of simple process and low cost. Based on the method, scientific releasing planning can be made to effectively promote recovery of wild Marsupenaeus Japonicus population resources, and filial generations of released individuals can also be detected. The method provided by the invention also can be used for years of continuous monitoring.

Description

technical field [0001] The invention belongs to the field of marine animal proliferation and release, and particularly relates to a method for evaluating the release effect of Penaeus japonicus. Background technique [0002] Japanese sac shrimp, also known as Japanese prawn (Kuruma prawn), scientific name Marsupenaeus japonicas, alias car shrimp, monodon shrimp, bamboo shrimp, taxonomically belonging to Crustacea, Decapoda, Swimming suborder, Penaeidae, Penaeus genus . Originating in Japan, China, the Philippines, Australia and other countries, it is widely distributed in India and the Western Pacific region, mainly in the Japanese archipelago, the Red Sea in South Africa, the Arabian Gulf, the Bay of Bengal, Japan and my country and other sea areas, with the largest number of coastal areas in Japan. The bottom of the habitat is mainly sandy and has the habit of burrowing sand. In natural sea areas, it burrows in the sand since the larvae stage, comes out for food at high t...
