Problems solved by technology

The main reason is that common ionic liquids and macromolecular crowding reagents are immiscible, so it is difficult to form a homogeneous system to complete the polymerization process, so it is necessary to develop a new porogen system to solve the problem of ionic liquids and macromolecular crowding agents. Miscibility of crowding reagents and good blotting results

Embodiment 1

[0025] Preparation of ursolic acid molecularly imprinted monolithic column using polymethyl methacrylate and ionic liquid as porogens and evaluation of imprinting effect:

[0026] a. The template molecule ursolic acid is added into a chloroform solution made of polymethyl methacrylate as a macromolecular crowding reagent, and 1-butyl-3-methyl-imidazole tetrafluoroborate and dimethyl sulfoxide as an ionic liquid. In a porogen with a volume ratio of 25:8:5, sonicate until the template is completely dissolved to form a complex;

Add functional monomer 4-vinylpyridine, cross-linking agent ethylene glycol dimethacrylate and initiator azobisisobutyronitrile to the compound formed in step a, degas it by ultrasonic for 10 minutes, and pass nitrogen gas for 10 minutes to remove Oxygen, and then inject the mixed solution into the stainless steel chromatographic column with a column length of 100 mm and an inner diameter of 4.6 mm. The molar ratio of the functional monomer an...

Abstract

The invention relates to a method for preparing a molecularly-imprinted monolithic column by using a molecular crowding reagent and ionic liquid as a pore-foaming agent. The method comprises the following steps: through special formulation of the pore-foaming agent, adding a template molecule ursolic acid or oleanolic acid into a pore-foaming agent consisting of a macromolecular crowding reagent, the ionic liquid and dimethyl sulfoxide, adding a functional monomer, a cross-linking agent and an initiator, performing ultrasonic degassing, and introducing nitrogen for removing oxygen to obtain an ursolic acid or oleanolic acidmolecularly-imprinted monolithic column. By the method, the macromolecular reagent and the green solvent ionic liquid can form the molecularly-imprinted monolithic column with good permeability and a significant imprinting effect; the imprinting effect of the obtained ursolic acid or oleanolic acidmolecularly-imprinted monolithic column is better than that when the macromolecular reagent and the ionic liquid are independently used as the pore-foaming agent; the method has the characteristics of simple preparation, avoidance of a complicated preparation process, good molecular recognition performance and the like; the obtained molecularly-imprinted monolithic column has good permeability and a significant imprinting effect; a new method is provided for preparing the molecularly-imprinted monolithic column with better imprinting effect.

Description

technical field [0001] The invention belongs to the field of pharmaceutical analysis and relates to the preparation of a molecularly imprinted monolithic column, in particular to a method for preparing a molecularly imprinted monolithic column by using macromolecular crowding reagents and ionic liquids as porogens. Background technique [0002] Molecular imprinting technology is a technology with pre-selective stationary phase developed on the basis of simulating the interaction of enzyme-substrate and receptor-antibody in nature. The imprinted polymer prepared by this technology has the advantages of good affinity and selectivity, strong resistance to harsh environments, good stability, long service life and wide application range. It has been widely used in chemical catalysis, solid phase extraction, chromatographic separation, Sustained and controlled release drug delivery systems, chemical sensors and other fields. [0003] In-situ polymerization is a kind of pre-polyme...
