Problems solved by technology

However, there are generally problems of sampling and detection difficulties: 1. The loss of cells and micro-nano particles is easy to be caused during the sampling process, which cannot accurately reflect the dose-effect relationship of micro-nano particles on cells; 2. Cells and micro-nano particles with low density When the particles are co-cultured, because the density of the micro-nano particles is lower than that of the liquid medium, the micro-nano particles float above the medium during the co-cultivation, and cannot directly contact the cells growing on the bottom of the culture plate, thus affecting the results of cell experiments
During the culture process, the cells are attached to the top (the bottom of the inverted culture plate), while the micro-nano particles will float above the medium, that is, they will gather at the bottom of the inverted culture plate. Although this method solves the problem of the contact between the micro-nano particles and the cells problems, but the operation is difficult, the medium is difficult to change, and long-term culture cannot be carried out; and when the separation medium is required to detect the morphology, proliferation, differentiation and cytokines of the cells and other related characteristics, the micro-nano particles are easy to be carried out at the same time, and will also be Test results that affect the relationship between micro-nano particles and cells

Abstract

The invention relates to a preparation method of a microreactor for experimenting influence of micro-nano particles on cells. The preparation method disclosed by the invention comprises the following steps of (A), sufficiently and uniformly mixing the cells and the micro-nano particles in polyanion isotonic solution to form a mixed solution; (B), dropping the mixed solution in a collecting tank containing divalent cation isotonic solution by using an electrostatic micro-injection machine to obtain a gel microsphere wrapped with the cells and the micro-nano particles; (C), putting the gel microsphere obtained in the step (B) in polycation isotonic solution to form a polycation wrapping film, then, putting the gel microsphere obtained in the step (B) in the polyanion isotonic solution and oscillating to form a polyanion wrapping film, and repeating the operation for 0-3 times to obtain the gel microsphere with 2-8 wrapping films; and (D), putting the gel microsphere obtained in the step (C) in divalent cation isotonic displacement solution so as to obtain the micro-reactor for experimenting influence of the micro-nano particles to the cells. According to the micro-reactor prepared by the invention, the cells and the micro-nano particles are contacted sufficiently; the cells and the micro-nano particles are accurate to measure, and thus, the influence relationship of the micro-nano particles on the cells can be reflected more accurately and reliably.

Description

technical field [0001] The invention relates to a preparation method of a micro-reactor used for experiments on the influence of micro-nano particles on cells. Background technique [0002] With the development of society and economy, environmental problems are becoming more and more serious. There are many micro-nano (micron, nano-scale) particles in the air, and the development of science and technology has revealed that PM2.5 (particles with a particle size less than or equal to 2.5 microns) in the environment seriously threatens human health. How PM2.5 micro-nano particles cause cytotoxicity and human pathological changes has become a common concern. In addition, the use of micro-nanoparticles to treat various diseases has also become a research and development hotspot. However, due to the large specific surface area and small volume of micro-nano particles, it is difficult to evaluate their influence on cells and the relationship between them. [0003] By placing the...
