Abstract
The invention relates to a nanoprobe capable of analysis and detection of activity of cancer marker MicroRNAs and MicroRNAs in cancer cells and a preparation method therefor. The nanoprobe is a novel four-helical DNA molecule probe. The nanoprobe contains three functional parts: a target object identification area loaded with special design, a trigger area triggering a cyclic amplification reaction and a template area for a polymerization-shearing amplification reaction. The nanoprobe is introduced into a surface enhanced Raman scattering (SERS) analysis detection technology, a four-helical molecule probe-SERS detection method is developed, and high-sensitivity detection of microRNAs can be achieved. Combined with the aptamer identification technology, the four-helical molecule probe-SERS detection method has good specificity, the sample treatment process is simplified, the selectivity is good, the accuracy is high, and the method is an ideal method for detecting the tumor cell marker microRNAs and microRNAs in cancer cells. The detection method and kit can distinguish cancer cells and normal cells, and has advantages of simple operation, and high-sensitivity and high-specificity detection of the cancer marker microRNAs, and thus the kit has good application prospects.
