Abstract

The invention discloses a breeding method of cortex eucommiae triploids. The breeding method comprises the steps of carrying out high-temperature processing on female flower buds of cortex eucommiae, and sequentially carrying out pollination, seed collection, sowing, seedling raising and ploidy detection, wherein the high-temperature processing is to processing the female flower buds of cortex eucommiae of different developmental stages at 42-48 DEG C after male plants at the peripheries of female plants of cortex eucommiae begin to release powder. According to the breeding method, parameters including processing period, processing temperature, continuous processing time and the like, which remarkably influence the yield of the cortex eucommiae triploids, of high temperature induced megaspore chromosome bred triploids are screened and optimized, so that the induction reliability of the cortex eucommiae triploids is remarkably improved, the induction efficiency of the cortex eucommiae triploids is obviously improved, the workload is greatly reduced, and the cost is effectively saved; and the breeding method has relatively strong operability.

Description

technical field [0001] The invention relates to a method for breeding eucommia triploids, in particular to a method for inducing eucommia megaspore chromosome doubling at high temperature to increase the yield of eucommia triploids, and belongs to the field of eucommia triploid breeding. Background technique [0002] Polyploid refers to individuals whose somatic cells contain three or more chromosome sets. Polyploidy exists widely in nature, and usually originates from chromosome doubling during mitosis of somatic cells or hybridization between unreduced gametes and normal gametes during meiosis. At present, the commonly used methods of artificially inducing chromosome doubling include chemical induction, physical induction and biotechnology. [0003] Eucommia ulmoides belongs to diploid (2n=2x=34), and it is a potential gum source tree species in temperate zone. It is also of great value in the utilization of medicinal ingredients such as iridoid glycosides. Judging from...
