Embodiment 1

[0053] 1. Acquisition of a set of kiwifruit species-associated specific single nucleotide molecular markers

[0054] Public sequence database resources: National Center for Biotechnology Information (NCBI for short), Kiwifruit Reference Genome Database (Kiwifruit Information Resource, KIR for short), internal database resources of National Kiwifruit Germplasm Resource Garden (Wuhan), the specific steps are as follows figure 1 shown, including:

[0055] ① Obtain the sequence sequence of kiwifruit species in the database

[0056] Using the Latin names of all species of Actinidia as keywords, search and obtain sequence data resources of more than ten Actinidia species in public sequence databases and internal databases.

[0057] ② Homologous sequence comparison to obtain preliminary variation information of species-associated specific single nucleotides

[0058] Use Cross Match software to remove useless sequences such as vectors from the obtained data sequences, use Gmap softw...

Abstract

The invention discloses kiwi fruit species association specific mononucleotide molecular markers and a detection primer set and application thereof. By means of sequencing sequences of different kiwi fruit species existing in a kiwi fruit genome database, high-diversity mononucleotide variation regions are looked for in a sequence alignment reference genome mode, a mononucleotide molecular marker primarily selected set is formed, on the basis, a novel mass spectrometer gene typing platform is used for carrying out experimental verification and genotype acquisition on mononucleotide variation sites, species association specific mononucleotide markers are screened through association analysis with the species, finally, a set of molecular markers for kiwi fruit hybrid resource parent species source identification are obtained, and the technical scheme is formed. Identification of kiwi fruit hybrid resource parents can promote understanding of formation of hybrid resource characters and biological characters and accelerate genetic evaluation and development and utilization of hybrid resources, and great theoretical academic significance and high economic application value are achieved.

Description

technical field [0001] The invention belongs to the field of plant genetic resource evaluation and biotechnology, and specifically relates to a set of kiwifruit species-associated specific single nucleotide molecular markers and its detection primer set and application, that is, it is used for the analysis and identification of the parent source of kiwifruit natural hybrid resources, so as to improve the quality of kiwifruit The screening and evaluation of hybrid germplasm and the utilization efficiency of genetic breeding can reduce land and labor costs and promote the rapid breeding of new varieties. Background technique [0002] Kiwifruit (Actinidia chinensis Planch.) is an important emerging fruit in the world originating in my country. It is widely loved by consumers because it is rich in vitamin C and other nutrients. Actinidia is rich in wild species resources, including 54 species and about 75 taxa. Due to the biological characteristics of dioecious and perennial ki...
