Embodiment 1

[0027] The composition for cartilage damage repair is prepared according to the following steps, and the cartilage damage patients are transplanted and repaired:

[0028] 1. Isolation and culture of UCMSC

[0029] The obtained fresh umbilical cords are sent to the GMP production workshop, and UCMSCs are obtained by separation. The UCMSCs were cultured and expanded; some cells were taken out to induce chondrocytes, and a certain number of chondrocytes were obtained and then expanded for transplantation.

[0030] 2. Preparation before transplantation

[0031] Digest the UCMSCs obtained above and the chondrocytes induced to differentiate with 0.25% trypsin, wash with saline, then mix the UCMSCs and chondrocytes at a ratio of 1:1 to 4:1 and resuspend them in hyaluronic acid solution to obtain a cell suspension, wherein the content of hyaluronic acid in the cell suspension is 5-30 mg / ml. The cell suspension is mixed with the temperature-sensitive polylactic acid hydrogel MPEG-PL...

Abstract

The invention discloses composition for cartilage injury repair. The composition is prepared from a cell suspension and temperature-sensitive type hydrogel, wherein the cell suspension is formed by mixing mesenchymal stem cells and cartilage cells and resuspending the mixture in a hyaluronic acid solution. The composition for cartilage injury repair is convenient to obtain, safe and reliable, and the composition differentiates towards cartilage cells efficiently and can provide cells essential to tissue repair for a cartilage injury part; the temperature-sensitive type hydrogel has very good functions of exogenous cell carriers, enables cells in the composition to fill the cartilage injury part sufficiently and prevents the cells from transferring to other places, and accordingly, the composition can effectively contribute to quick and efficient repair and healing of the cartilage injury part.

Description

technical field [0001] The present invention relates to compositions for repairing cartilage damage. Background technique [0002] Articular cartilage (AC) damage is a major risk factor for osteoarthritis (OA). OA is irreversible. When the disease develops to an advanced stage, joint replacement is required to relieve pain and restore function. Since the repair capacity of articular cartilage is limited, early intervention is necessary to prevent progression to OA. Current treatment modalities aim to repair articular cartilage through primary therapy, stimulation of adjacent tissues, and grafting. Reasonable restorative treatment can achieve ideal results in the short to medium term, but after a long period of time, it is easy to relapse. In recent years, tissue engineering technology based on bioengineered tissue at the cell level has been extensively studied. This bioengineered tissue can reproduce the characteristics of hyaline cartilage and integrate with the original ...
