Problems solved by technology

In the future, there will be a large demand for iPSCs-induced nerve cells in both the clinical treatment field and the precision medicine field. At present, there are many methods for generating nerve cells from stem cells, most of which are only done in the laboratory, and the differentiation efficiency of nerve cells is low. High, the difference between cell lines is very large, it is not conducive to large-scale production and application, and cannot meet the large needs of safe, homogeneous, precision medical services and drug testing industries

Embodiment Construction

[0026] In order to better illustrate the purpose, technical solutions and advantages of the present invention, the present invention will be further described below in conjunction with the accompanying drawings and specific embodiments. Unless otherwise specified, the technical means used in the examples are conventional means well known to those skilled in the art, and the raw materials used are commercially available items.

[0027] As used herein, the term "induced pluripotent stem cell", often abbreviated as iPS cell, refers to an adult that has been reprogrammed to enter an embryonic stem cell-like state by being forced to express factors important for maintaining the "sternness" of embryonic stem cells cell. Generally, iPS cells are artificially prepared from non-pluripotent cells (such as adult human cells, or terminally differentiated cells), such as fibroblasts, hematopoietic cells, muscle cells, neurons, epidermal cells, etc., by The non-pluripotent cells are introd...

Abstract

The invention provides a kit and a method for in-vitro directionally differentiating human-derived iPS stem cells into nerve cells. By utilizing a culture solution provided by the kit and according to the method, the iPS stem cells can be efficiently and directionally differentiated into the nerve cells. The kit and the method are simple, reliable, stable, efficient and high in safety, proportion of differentiating pluripotent stem cells into nerve cells is obviously increased, and differentiating proportion of the pluripotent stem cells is higher than 90%. In addition, the kit and a culture medium are suitable for large-scale production of high-quality nerve cells and can be directly used for scientific research on neurodevelopment, cell therapy of nerve diseases, nerve injury repair and application of drug screening without subsequent screening and purifying.

Description

technical field [0001] The invention relates to the fields of stem cell biology and regenerative medicine, in particular to a kit and method for directional differentiation of human iPS stem cells into nerve cells in vitro. Background technique [0002] Induced pluripotent stem cells (iPSCs) were successfully created by the Yamanaka research group in Japan in 2006 through reprogramming technology. Subsequently, many laboratories at home and abroad used transgenic methods to reprogram various types of adult cells into iPSCs. iPSCs introduce certain transcription factors into animal or human somatic cells through gene transfection technology, so that somatic cells can be directly reprogrammed into embryonic stem cell (embryonic stem cell, ES) cell-like pluripotent cells. iPS cells are not only very similar to ES cells in terms of cell morphology, growth characteristics, and expression of stem cell markers, but also almost completely similar to ES cells in terms of DNA methyla...
