Embodiment 1

[0039] Example 1 High-throughput sequencing screening of differentially expressed genes

[0040] 1. Sampling

[0041] Patients with osteoporosis who were treated in the Department of Orthopedics of Peking Union Medical College Hospital from October 2012 to December 2015 were selected. A total of 26 cases were collected in the case group. All patients had typical clinical manifestations. Diagnosed. Controls were collected from patients with other diseases who were hospitalized in orthopedics at the same period, and a total of 22 cases were collected. Blood samples of all subjects were collected and numbered and stored in a -80°C freezer. All clinical samples in this study were informed to the patients and approved by the Hospital Ethics Committee.

[0042] 2. Total RNA extraction from blood samples

[0043] Trizol LS (Invitrogen: 10296-010) was used to extract RNA from the collected blood samples, and the experimental operation was carried out according to the product instr...

Embodiment 2

[0054] Example 2 RT-PCR verification of ZNF800 gene expression in the blood of osteoporosis patients and controls

[0055] 1. Materials

[0056] A total of 10 patients with osteoporosis who visited the Department of Orthopedics of Peking Union Medical College Hospital from October 2012 to December 2015 were selected. All patients had typical clinical manifestations and were confirmed by bone density (dual-energy X-ray) examination. Controls came from patients with other diseases who were hospitalized in orthopedics at the same period, and a total of 8 cases were collected. Blood samples of all subjects were collected and numbered and stored in a -80°C freezer. All clinical samples in this study were informed to the patients and approved by the Hospital Ethics Committee.

[0057] 2. Method

[0058] 2.1 Total RNA was extracted from the blood sample, which was the same as the extraction method in Example 1.

[0059] 2.2 Synthesis of cDNA by reverse transcription

Embodiment 3WB

[0080] Example 3WB method detects the protein expression level of ZNF800 in the blood of patients with osteoporosis

[0081] 1. Materials

[0082] Collected 10 cases of osteoporosis patients who underwent kyphoplasty surgery, and obtained bone tissue from diseased vertebrae of osteoporosis patients during the operation. The samples were collected from the orthopedic department of Peking Union Medical College Hospital from October 2012 to December 2015. All patients with osteoporosis had typical clinical manifestations and were confirmed by bone density (dual-energy X-ray) examination. Controls were obtained from patients with spinal burst fractures caused by trauma who were hospitalized at the same time. A total of 8 cases were collected, and normal vertebral bone tissues were obtained. Bone tissue samples from all subjects were collected and numbered and stored in a -80°C freezer. All clinical samples in this study were informed to the patients and approved by the Hospital ...

Abstract

The invention discloses a molecular marker for early osteoporosis screening. The molecular marker is a ZNF800 gene or ZNF800 gene expression protein and a fragment, analogue and derivative thereof. The invention further discloses application of the ZNF800 gene in preparation of an early osteoporosis screening product. By conducting general investigation on osteoporosis in the crowd by means of the gene, not only can early osteoporosis screening be quickly and effectively achieved, but also a treatment target and an important basis are provided for prediction and diagnosis on occurrence and development of the osteoporosis and even for osteoporosis treatment in the biological level in future.

Description

technical field [0001] The invention relates to the field of molecular biology, in particular to the application of the ZNF800 gene in the preparation of early screening products for osteoporosis. Background technique [0002] Osteoporosis is a metabolic bone disease characterized by decreased bone mass and microarchitectural destruction of bone tissue, resulting in increased bone fragility and susceptibility to fracture. Osteoporosis has a slow onset and is characterized by bone pain and fragility fractures. With social development and population aging, osteoporosis has become one of the most common diseases. At present, osteoporosis has become a frequently-occurring and common disease, and the incidence rate increases with age. my country is the country with the largest population and elderly population in the world, and it is also the country with the largest number of patients with osteoporosis and potential osteoporosis. In my country, there are about 29 million pati...
