Abstract

The invention relates to a targeted platelet membrane glycoprotein GPIbalpha monoclonal antibody for inhibiting tumor metastasis. The monoclonal antibody is a monoclonal antibody that the specificity thereof is combined with a platelet membrane protein GPIbalpha. The invention also relates to a method for screening a tumor metastasis inhibitor of a targeted platelet membrane glycoprotein GPIbalpha. The method comprises five steps of: 1, establishing an initial screening model by using an enzyme-linked immunosorbent assay; 2, establishing a secondary screening model with Western blotting; 3-5, establishing a functional detection model with a flow type cytometer and a continuous spectrum luminoscope. The established screening model significantly reduces the need for human-derived platelets during the process of acquiring the targeted platelet membrane glycoprotein GPIbalpha inhibitor and maximizes the activation of tumor cells in a vitro experiment, simulates the microenvironment of the tumor cells and improves the screening accuracy, and the obtained antibody YQ3 has a good clinical application prospect.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to pharmacology, and specifically refers to a monoclonal antibody targeting platelet membrane glycoprotein GPIbα for inhibiting tumor metastasis and a screening method thereof. Background technique [0002] Platelets can promote tumor metastasis, and the first and key point in promoting tumor metastasis is the activation of platelets by tumor cells to form tumor thrombi. A variety of adhesion molecules on the surface of platelets, such as glycoprotein (Glycoprotein, GP) Ib-IX-V, GPIIb-IIIa, interact to mediate the adhesion of tumor cells to platelets. Among them, GPIbα is the largest and most important component of the GPIb-IX-V complex. Studies have shown that knocking out the GPIbα gene or replacing the GPIbα extracellular region in mouse models can significantly reduce the lung metastasis of lung cancer in mice, but it will be complicated by thrombocytopenia (Jain S, Zuka M, Liu J, ...
