Problems solved by technology

[0003] In the artificially collected giant panda semen, the active sperm will gradually lose their activity in a normal temperature environment, which will affect the conception rate of artificial insemination
[0004] After giant panda semen is collected, it needs to be frozen and transported. If it is not handled properly, it will lead to a decline in sperm motility, which will greatly reduce the success rate of artificial insemination.

Embodiment 1

[0042] (1) The semen of male giant pandas aged 5.5-20.5 years old was collected by electric stimulation method, and the sperm motility was checked. If the motility was greater than 80%, it was qualified.

[0043] (2) Prepare the diluent: the diluent uses the Test Yolk Buffer with Gentamicin diluent from Irvine Scientific Company.

[0044] (3) Dilute qualified semen for the first time in a 15ml centrifuge tube with an isothermal diluent at a ratio of 1:1 in a foam incubator at 30-33°C, and then place it in a 250ml beaker. Equilibrate in a refrigerator at 4°C for 3.5 hours, and then add antifreeze protectant. Glycerin is used as the antifreeze protectant, and the ratio of the volume of glycerin to giant panda semen is 1:1. Continue to place it in a 4°C refrigerator for 1 hour, then put the semen into a 0.25ml seminal tube and place it in a dry place in a 4°C refrigerator. (When performing the above steps, open the refrigerator door, and the speed should be fast to ensure the st...

Abstract

The invention discloses a dry-tank freezing method of giant panda seminal fluid. The dry-tank freezing method comprises the following steps: (1) diluting collected giant panda seminal fluid through a diluent in 30 to 33 DEG C; (2) putting the diluted giant panda seminal fluid into 4 DEG C environment to be balanced; (3) adding an antifreeze protective agent into the balanced giant panda seminal fluid and continuously putting in the 4 DEG C environment; (4) transferring the giant panda seminal fluid treated in the step (3) into a container, covering the container in a gauze bag, putting the gauze bag into a dry liquid nitrogen tank, putting the gauze bag at the position 6cm under a tank opening of the dry liquid nitrogen tank, capping and staying for 5min, then putting the gauze bag at the tank bottom of the dry liquid nitrogen tank, sealing and transporting; (5) putting into a general liquid nitrogen tank to be stored after transportation finishes and taking out to be unfrozen in 50 DEG C hot water for 10s when the giant panda seminal fluid needs to be used; or unfreezing for 10s in 50 DEG C hot water to be used after transportation finishes. The method is convenient to operate, is suitable for national parks or field places and has important significance in heredity value storage of wild animals; furthermore, utilized equipment is simple.

Description

technical field [0001] The invention belongs to the field of sperm preservation, and in particular relates to a method for freezing giant panda semen in a dry tank. Background technique [0002] The giant panda is a rare species unique to my country, with low reproductive ability and short estrus period. In the captive giant panda population, due to the small number of male and female giant pandas in the breeding season at the same time, the success rate of natural mating is low, so artificial insemination is sometimes used to promote the pregnancy of giant pandas. [0003] The active sperm in the artificially collected giant panda semen will gradually lose their activity in a normal temperature environment, which will affect the conception rate of artificial insemination. [0004] After the giant panda semen is collected, it needs to be frozen for storage and transportation. If it is not handled properly, it will lead to a decline in sperm motility, which will greatly redu...
