Embodiment 1

[0019] Example 1: Noise exposure down-regulates the expression and dissimilation of glutamate receptor protein in the A1 and MGB regions. Its subtissue distribution is firstly fixed by perfusion: after the experimental animals are weighed, they are given an excessive intraperitoneal injection of 10% chloral hydrate. anaesthetization. After entering the anesthesia state, the chest cavity was opened, and about 200ml of cold normal saline was perfused from the left ventricle by infusion until the perfusate of the right atrium became clear. After that, 4% paraformaldehyde fixative was used to replace the normal saline for 30 minutes. Left and right, and finally the whole brain was taken and fixed in 4% paraformaldehyde fixative solution. Next, the tissue blocks are immersed in 20% and 30% sucrose solutions in turn until they sink to the bottom; then sliced: the dehydrated tissue blocks are embedded in OCT, quickly frozen, and then placed on the sample holder, and serial coronal cr...

Embodiment 2

[0022] Example 2: Noise exposure down-regulates the expression of glutamate receptor mRNA in A1 and MGB regions and its subtissue distribution According to the target ion channel sequence found in NCBI, qRT-PCR primers were designed (see Table 1.1). The primer design follows the principle that the primer length is about 20bp and the product size is about 100bp, and is synthesized by Nanjing GenScript Biotechnology Co., Ltd. Primer sequences are shown in Table1.1. The PCR amplification products of all primers were verified by sequencing. qRT-PCR uses the AceQ qPCRSYBR Green Master Mix kit, and the reaction system is as follows:

[0023] Reaction system components (20μL): AceQ qPCR SYBR Green Master Mix 10.0(μL), Primer 1(10mM) 0.2(μL), Primer 2(10mM) 0.2(μL), cDNA 1.0(μL), ddH20Up to 20(μL)

[0024] The reaction conditions are as follows:

[0025] Steps: pre-denaturation at 95°C, 5s, 1 cycle; denaturation at 95°C, 10s, 30 cycles; annealing extension at 60°C, 30s, 3 cycles; m...

Abstract

The invention discloses the application of an ionotropic glutamate receptor in the preparation of medicine for treating noise-induced hearing loss. Experiments showed that the protein expression levels of GluR1, GluR2, NR2A, NR2B, and NR1 receptors decreased in different degrees in A1 and MGB regions, and their mRNA expression levels also decreased in different degrees. Among them, the down-regulation of NR2A receptor protein expression and mRNA expression level is particularly obvious, and it should be an ideal new drug target for the treatment of noise-induced hearing loss.

Description

technical field [0001] The present invention relates to the use of an ionotropic glutamate receptor. Background technique [0002] Noise induced hearing loss (NIHL) has become one of the common occupational diseases. Noise-induced auditory system damage is first concentrated in cochlear hair cells and primary afferent neurons. Long-term exposure to noise can cause short-term or long-term hearing threshold dissimilation and other cochlear lesions, and even induce permanent hearing loss. Noise exposure disrupts the coding of peripheral auditory input into the cochlea and its output into the central auditory system. Excessive noise stimulation can lead to damage or death of the outer hair cells and inner hair cells in the cochlea, and alienate the incoming and outgoing sound signals. Noise stimulation can also change the spectral distribution of the cochlea and weaken the audio frequency perception ability of the auditory center. Then change the physiological activity of neu...
