Problems solved by technology

Cells of the innate system recognize and respond to pathogens in a universal manner, but unlike the adaptive immune system, the innate system does not confer long-lasting or protective immunity to the host

Embodiment 1

[0087] Example 1: Dose Response Effect of Turkic Rose and Poria Cocos Extracts on Phagocytosis

[0088]THP-1 monocytes were treated with 1 nM phorbol 12-myristate 13-acetate (PMA), and extracts of Turkic rose and Poria cocos at the indicated concentrations for 20 hours. Cells were labeled with Cell TrackerGreen, washed, and transferred to 5 mL polypropylene tubes and CypHer5e-labeled 3 mm latex beads and incubated for an additional 18 hours. After incubation, cells were washed, resuspended in ice-cold PBS, and analyzed by flow cytometry using FL1 (Cell Tracker) and FL4 (CypHer). Cypher only fluoresces in the acidic environment of the phagosome. Data were analyzed by establishing regions of FL1+ cells (R2) and FL1+, FL4+ cells (R3). R3:R2 ratios were determined for each experimental condition and each ratio was divided by the R3:R2 ratio calculated for cells treated with PMA only to generate a phagocytic index (findex).

[0089] The phagocytosis method is based on Beletski A...

Embodiment 2

[0091] Example 2: The effect of the combination of Turkic Rose and Poria cocos extracts on phagocytosis.

[0092] THP-1 cells were treated with PMA, rose extract (50 mg / mL) alone or in combination with Poria cocos extract (5 or 10 mg / mL) for 20 hours.

[0093] Phagocytosis was assessed as described in Example 1 above.

[0094] Such as figure 2 As shown, when the cells were treated with the combination of Turkic Rose and Poria extracts at a ratio of 5:1 or 10:1, the level of phagocytosis showed an unexpected and surprising increase, which was higher than when treated with either alone. The extract will respond at the expected level.

Embodiment 3

[0095] Example 3: Effect of Turkic rose and Poria cocos extracts on phagocytosis of zymosan A, heat-inactivated yeast.

[0096] Will 1X 10 5 THP-1 cells were plated in 12-well glass-bottom plates (No. 1.5) for 48 hours. Add 1 nM PMA and fresh growth medium for 24 hours. Cells were treated with Turkic Rose, Poria cocos or a mixture for 5 hours. A mixture of Zymosan A (Texas Red Conjugate) and Opsonizing Reagent was added to each well to generate 500,000 Zymosan A / well for 1 hour. Cells were fixed with 4% formaldehyde in PBS, stained with Acti-Ready 488 for actin, and mounted in Diamond Anti-Fade Mountant with DAPI nuclear stain.

[0097] Fluorescence was captured on a Nikon Eclipse Tifluorescent microscope and Photoshop was used to sharpen images and generate graphics.

[0098] Visualization of enhanced phagocytosis by Turkic rose and Poria cocos extracts by fluorescence microscopy ( Figures 3A to 3D ). Although it was difficult to show quantitative differences between t...

Abstract

A composition comprising a Rosa Damascena component or a Rosa Damascenaextract and a Poria cocoscomponent or a Poria cocosextract is disclosed. A method using the composition to activate and / or enhance an immune response in a subject is also disclosed.

Description

technical field [0001] In general, the present disclosure relates to compositions and methods for activating and enhancing a subject's immune system and / or response. Background technique [0002] The immune system is the host defense system comprising many biological structures and processes in an organism that provide protection against disease. To function properly, the immune system must detect an enormous variety of agents (called pathogens), ranging from viruses to parasitic worms, and distinguish them from the organism's own healthy tissue. [0003] In many species, the immune system can be divided into subsystems, such as the innate and adaptive immune systems, or humoral and cell-mediated immunity. [0004] Disorders of the immune system can lead to autoimmune diseases, inflammatory diseases and cancer. Immunodeficiency occurs when the immune system is less active than normal, leading to recurrent and life-threatening infections. In humans, immunodeficiency can be...
