[0044] 1. Preparation of BMP-2 sustained release system mediated by PLGA microcapsule scaffold

[0045] (1) Preparation of PLGA microcapsules

[0046]Dissolve BMP-2 in 200 μg of distilled water. In order to ensure full dissolution, a magnetic stirrer can be used to stir for 5 minutes to prepare W1 solutions with different BMP-2 concentrations; 2 Cl 2 and TFE (tetrafluoroethylene) at a volume ratio of 1:4) to obtain an organic phase O; mix the W1 solution with the organic phase evenly, and ultrasonically emulsify for 20 minutes to obtain an initial emulsion; dissolve polyvinyl alcohol (PVA) In 40mL of water, a W2 solution with a PVA concentration of 5% was obtained as the external water phase; the initial emulsion was added dropwise to the external water phase, and ultrasonically emulsified for 10min to form a double emulsion; stirred at room temperature for 2h at a speed of 800rpm to make the organic solvent Volatilize to solidify the microcapsules; centrifuge the microcapsu...

Abstract

The invention discloses application of a combination of a PLGA microcapsule stent encapsulated with BMP-2 and PRF in preparation of drugs for promoting tendon-bone healing. According to the invention, the slow release of BMP-2 of the PLGA microcapsule stent and PRF implantation are combined, wherein BMP-2 released slowly can promote the formation of bone cells and chondrocytes at a tendon-bone healing interface at a relatively uniform speed, and cytokines are slowly released by PRF can promote vascularization at the tendon-bone healing interface at a relatively uniform speed, so as to promotethe healing of a bone-tendon junction. BMP-2 and PRF promote each other synergistically, and PRF can effectively promote the healing ability of the new bone cells and chondrocytes and the bone-tendonjunction, and only when the new bone cells and chondrocytes and the bone-tendon junction heal well, BMP-2 can further promote the effective regeneration of bone cells and chondrocytes.

Description

technical field [0001] The invention belongs to the technical field of tendon-bone healing after ligament reconstruction, and in particular relates to the application of PLGA microcapsule scaffolds encapsulated with BMP-2 combined with PRF in the preparation of drugs for promoting tendon-bone healing. Background technique [0002] The anterior cruciate ligament (ACL) is an important static and stable structure of the knee joint. It participates in the maintenance of frontal and lateral rotational stability and is extremely important for maintaining the function of the knee joint. Due to the extremely poor self-repair ability after ACL injury, the main method of clinical treatment is surgical reconstruction. It is estimated that there are more than 200,000 ACL reconstruction operations in the United States every year (Frobell RB, Roos EM, RoosHP.et a1.A randomized trial of treatment for acute anterior cruciate ligamenttears. N Engl J Med, 2010, 363: 331-342.). Most of the re...
